The Study On The Processing Standardization Of Nelumbo Nucifera Gaertn Pieces

Objective:To optimize the processing methodology of Nelumbo nucifera Gaertn Pieces, to formulate the quality standards for Nelumbo nucifera Gaertn Pieces, to study the changing law of the main active ingredients of Nelumbo nucifera Gaertn produced before and after, and discuss the processing principle.Methods:On the basis of ancient and modern literature of Nelumbo nucifera Gaertn produced, the contents of the main active ingredients were taken as indexes and the orthogonal design method was used to carry out the filtration and optimization of the processing methodology of Nelumbo nucifera Gaertn pieces(Nelumbo nucifera Gaertn, carbo Nelumbo nucifera Gaertn) which were collected in "Chinese Pharmacopoeia" and "Standard of National Chinese Medicine Concoction", determined the specific technical parameters of the process; Prepared each 10 batch samples of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn by optimized processing methodology of Nelumbo nucifera Gaertn pieces, identified the characters and TLC, established the corresponding identification methods, checked each 10 batch samples of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn'impurities, moisture, total ash, acid insoluble ash and then restrict 120% of 95% confidence interval of every index and component average content as their content upper limit combined with the actual production; Mensurated 10 batch samples of Nelumbo nucifera Gaertn'alcohol-soluble extract, total alkaloids and nuciferine, Mensurated 10 batch samples of carbo Nelumbo nucifera Gaertn' alcohol-soluble extract and quercetin, and then restrict 80% of 95% confidence interval of every index and component average content as their content lower limit combined with the actual production, inspected on the stability, studied and formulated quality standards of Nelumbo nucifera Gaertn and carbo Nelumbo nucifera Gaertn pieces.Result:After Nelumbo nucifera Gaertn was calcineded and parched, the total alkaloids and nuciferine was nearly loss, the content of tannin was slightly loss, while the quercetin content of more health products increased by 6 times.the processing methodology of Nelumbo nucifera. Gaertn pieces is:To take 100g hand-picked medical material, quick rinse, set the container, add the weight of 30% of the medicinal water moistening, turned over once every 30min, run 3h to do drug permeable, soft cut should, in time out, a little dry, cut 5～6cm wire,60℃drying, screened out debris, sealed in plastic bag and got it. The best condition of carbo Nelumbo nucifera Gaertn pieces was determined:set the temperature of drug speculation machine temperature control instrument as A℃and rotation speed at 20 r·min-1, took clean Nelumbo nucifera Gaertn pieces, fried 15 minutes till the supine surface become cinnamomeous and the superficies inferia become pyro-brown in 270℃, slight focal, took out for a while to dry, screened out debris, blended and sealed in separate plastic bags and got it. Nelumbo nucifera Gaertn pieces contained impurities, moisture, total ash and acid insoluble ash content which should be no more than 2.0%,13.0%,6.0% and 0.6%;70% ethanol-soluble extract, nuciferine and total alkaloids contents which should be no less than 15.0%,0.20%, and 0.60%. Carbo Nelumbo nucifera Gaertn pieces contained impurities, moisture, total ash, acid insoluble ash content which should be no more than 3.0%,14.0%,6.0% and 0.6%; 70% ethanol-soluble extract and quercetin content which should be no less than 13.0% and 0.40%.Conclusions:Comparing systematic content of the main chemical components Between the different processed products of lotus leaves, established the foundation for further research on the Nelumbo nucifera Gaertn processing mechanism and guidance for clinical medicine.The study identified the standardized production process and process parameters, instituted the quality standards for Nelumbo nucifera Gaertn pieces.
