Objectives observing the antagonistic effect of Dicliptera chinensis (L.)Ness polysaccharides in dimethylnitrosamine(DMN)-induced rat liver fibrosis,as well as TGF-β1 expression, it is provide a new way of treatment to research and develope Chinese medicine and the effective monomer for clinical anti-hepatic-fibrosis.Methods 80 Wistar rats were randomly divided into normal group, model group, Dicliptera chinensis polysaccharide high-dose group, low-dose group and the colchicine group a total of five groups of 16 rats, evenly divided male and female.1st day in laboratories, in addition to the normal control group than the other rats in each group are 0.5% DMN solution according to the weight 1.6ml/kg dose of intraperitoneal injection per week for 3 consecutive days, day 1, the first 1 week with 2 / 3dose, with the full amount of the future.Dicliptera chinensis polysaccharide high-dose (0.3g/kg), low-dose (0.1 g / kg) and colchicine group (0.1mg/kg) were gavage once a day, with normal saline to a total of 4 weeks.4 weeks killing the rats to get eye blood ,weighing the liver, spleen and liver tissue wet weight and charge; automatic biochemical analyzer determination of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total bilirubin (TBIL) and whiteprotein (ALB) Indicators; enzyme-linked immunosorbent assay (ELISA) detection of serum hyaluronic acid (HA), laminin (LN),Ⅲprocollagen (PCⅢ),Ⅲcollagen (Ⅲ-C); hematoxylin1 eosin staining (HE staining), and Masson staining to observe the formation of HF; immunohistochemical detection of transforming growth factorβ1 (transforming growth factorβ1, TGF-β1).Results①liver: Dicliptera chinensis polysaccharide high-and low-dose group of ALT, AST, TBIL, ALB were significantly lower than the model group, the differences were statistically significant (P <0.01).②serum indices of hepatic fibrosis: Dicliptera chinensis high-and low-dose of serum liver fibrosis four indicators (HA103.5±8.67 ng/mL,LN87.3±12.51ng/mL,PCⅢ30.8±3.68 ng/mL and IV-C49.7±4.56ng/mL) slightly higher than the normal group (P <0.05); but with the model group (HA182.4±16.62ng/mL,LN142.6±18.82ng/mL,PCⅢ56.3±4.91ng/mL,IV-C88.9±8.74ng/mL) compared to the dog liver vegetables high and low serum levels of four liver fibrosis were significantly lower (P <0.05, P <0.05, P> 0.05, P <0.05).③pathology HE and Masson staining: Dicliptera chinensis polysaccharide high-and low-dose group liver tissue necrosis and fibrosis in inflammatory lesions is higher than the model group significantly reduced, the differences were statistically significant (P <0.01).④immunohistochemical staining in liver tissue: model group, TGF-β1 expressed mainly in periportal areas and fiber spacing, and the elongated oval or spindle-shaped, immunohistochemical staining area was significantly higher than the normal control group.Dicliptera chinensis polysaccharide high and low dose TGF-β1 expression in model group decreased significantly compared with the normal control group, no significant difference.Conclusion 1.Dicliptera chinensis (L.)Ness polysaccharides with anti-liver DMN-induced liver fibrosis and its mechanism of liver protection and down-JiangMei liver TGF-β1 expression.2. Dicliptera chinensis (L.)Ness polysaccharide may be by down-regulation of the expression of TGF-β1, thereby inhibiting the activation of HSC, eventually leading to the synthesis of ECM degradation and reduction of HA secretion to play its anti-fibrosis.
