Objective: To study the impact of Dicliptera chinensis polysaccharideon hepatic fibrosis (HF) and the activation of hepatic stellate cells (HSC).Methods:1Construct the model by dimethyl nitrosamines (DMN) liver fibrosis inrat. HE and Masson staining evaluate the degree of HF.2The HF rats were treated with different concentrations of Diclipterachinensis polysaccharide (300mg/kg,100mg/kg). The expression ofα-SMA,TGF-β1and pSmad2/3protein were analysised by immunohistochemical.3HSC were activated by TGF-β1(1ng/mL). MTT assay was used todetermine the proliferation of HSC and then treated with differentconcentrations Dicliptera chinensis polysaccharide.4Inverted phase contrast microscope was used to observe themorphogical changes of the activated HSC.5Western Blotting was used to detect the expression of α-SMA, TGF-β1and pSmad2/3. Results:1Necrosis and fibrosis reduced dramatically through Dicliptera chinensispolysaccharide, inflammatory lesion is higherly reduced than the model groupsignificantly, Dicliptera chinensis polysaccharide (300mg/kg) especically.2After treated with Dicliptera chinensis polysaccharide, the espressionof TGF-β1, α-SMA, pSmad2/3decreased significantly, Dicliptera chinensispolysaccharide (300mg/kg) especically.3Dicliptera chinensis polysaccharide has inhibited the proliferation ofthe HSC in dose dependent way. But Dicliptera chinensis polysaccharide has noteffect on non-activted HSC (P<0.05).4The inverted phase contrast microscope: The activated HSC havemany microvillus in surface and looks like fibroblast.5With the increase of drug concentration, the expression of α-SMAand pSmad2/3protein decreased significently in activated HSC (P<0.05).Conclusions: Dicliptera chinensis polysaccharide inhibited theproliferation of the HSC, down-regulated the espression of α-SMA andpSmad2/3protein. Then play a key role in anti-hepatic fibrosis.
