Aim: To observe the effects of osteoprotegerin(OPG) on the change in number of endothelial cells and nitric oxide synthase (eNOS) , and to explore its significance to influence the proliferation of the endothelial cells.Method: The conventional culture of immortalized human endothelial cells, with different concentrations OPG (0,0.5,2.0,4.0,6.0,8.0 ug/L) to cultivate cells 48h, then with the cell count kit (CCK-8) to detect the number of cells, immunocytochemical method to detect eNOS protein expression, real-time quantitative PCR to detect the level of eNOS mRNA. Result: Compared with the control group (0.936±0.146), the cell number of 8.0 ug/L group (1.291±0.200)was evidently elevated (P <0.05); compared with the control group, eNOS protein expression in 2.0 ug/L, 4.0 ug/L group (P <0.05) increased significantly, and that increased more remarkably in 6.0 ug/L, 8.0 ug/L groups(P <0.01); in 4.0 ug/L, 6.0 ug/L, 8.0 ug/L groups eNOS mRNA were significantly increased (P <0.01) compared to the control group as well.Conclusion: The concentrations of OPG in humans range promoted endothelial cell proliferation. As the anti-atherosclerosis OPG may promote endothelial cells'repairation, and had evidently positive correlation with the concentration gradient.
