【Objective】: To observe the triptolide how to influence the rat's joint tissue which is with lesions of adjuvant arthritis and the expression of VEGF,FLT1 and CCR5 proteins. To explore the inhibition of the drug on the arthritis synovial tissue vascular proliferation and the molecular mechanism of inflammatory cell infiltration.【Methods】: On the basis of the rat model with adjuvant arthritis are given treatment by 0.1-0.4mg/kg triptolide and then measured the swelling of the lesion joint. Using the routine HE staining technical to detect the degree of inflammation; immunohistochemistry staining the joint tissue lesions to detect VEGF,FLT1 and CCR5 proteins expression. To analysis the content of FLT1 mRNA leaves by in situ hybridization. Using RT-PCR to detect the VEGF mRNA levels in disease.【Results】: Compared with model group,the joint swelling degrees of the other three triptolide treatment groups are significantly reduced;The pathological damage to joint tissue lesions are significantly improved; The number of the pathological joint tissue VEGF,FLT1 and CCR5 positive cells significantly reduced. The FLT1 and VEGF mRNA levels in lesions joint organization is decreased significantly.【Conclusion】: Triptolide could inhibit adjuvant arthritis rats with lesions of joint tissue in the expression of VEGF,FLT1 and CCR5,which may be the molecular mechanism of Triptolide inhibits proliferation of vascular lesions joints and the inflammatory cell infiltration.
