Radix sophorae flavescentis is the dried root of Sophora flavescens Ait. of pulse family Sophora plants. In this thesis, digitized fingerprints quality control methods of Radix sophorae flavescentis were established by HPLC and HPCE, and its super-information characteristic digitized evaluations were presented. The CZE method was also developed for the quantitative determination of matrine and oxymatrine in Radix sophorae flavescentis. Meanwhile, a software which named "The plane chromatogram fingerprint imaging system" was explored. This study provided new methods for overall quality control of Radix sophorae flavescentis and offered basis for realizing the quantitative function and comprehensive evaluation of fingerprints.1. The index of chromatographic fingerprints (F) was used as quantitative evaluation parameter to choose determining conditions. The HPLC digitized fingerprint of Radix sophoraeflavescentis under 220 nm was established and 22 common peaks were defined, taking the peak of oxymatrine as referential peak; the HPLC digitized fingerprint of Radix sophorae flavescentis under 265 nm was established and 35 common peaks were defined, taking the peak of kushenol 0 as referential peak. The qualitative similarities of crude drugs from different resources were evaluated by the software of Digitized Evaluation System for Super-Information Characteristics of Traditional Chinese Medicine Fingerprints 3.0 while the unified fingerprints of Radix sophorae flavescentis were established.2. The HPCE digitized fingerprints of Radix sophorae flavescentis were established by MECC and CZE methods, and evaluated by the software of Digitized Evaluation System for Super-Information Characteristics of Traditional Chinese Medicine Fingerprints 3.0 while the influences of different background electrolytes. pHs and voltages on the electropherogram were investigated. The CZE method was developed for the quantitative determination of matrine and oxymatrine in Radix sophorae flavescentis. The characteristics and practicability of Jaccard similarity S_b were investigated by comparing with the dual qualitative similarity parameters (both S_F and S_F') and the dual quantitative similarity parameters (both C and P). 3,"The plane chromatogram fingerprint imaging system" software was explored according to the theory of digital image processing technology with application of VB.NET 2003 based on Access database. It can more quickly solve the qualitative and quantitative problem of thin layer chromatography. This novel software was proved to be applicable to imaging formation and assessment of thin layer chromatography. And it was also validated by TLC experiment.
