| Objective:To observe the effect of the oil of jiaohao on the histology of testis, the level of plasma INHB and FSH of experimental autoimmune orchitis ( EAO) in mouse, and to explore its antagonism and it s mechanism. Methods: Draw Jiaohao oil in the first time, Experiments were performed on 180 healthy adult and male BALB/c mouses, weighting about 20-25 g, divide into 6 groups casually, and each group should divide to three time groups, each group has 10 mouses. The first group was lavaged by sodium chloride the second group only operation, the third group was lavaged by Olive oil, the forth group was lavaged by low dose Jiaohao oil, the fifth group was lavaged by midst dose Jiaohao oil, the sixth group was lavaged by high dose Jiaohao oil. Every group were lavaged before three days when they were made for operations, and go on lavaged after being operations. The experiments were observed in plan. To observe the change of testiculus seminiferous tubule by HE staining, and the change of FSH and INHB were detection by ELISA. Results: HE staining ,non–operation side:The three weeks time, in Sodium Chloride group, there is no spermatogenic cell amotic in mouses'seminiferous tubule, every classes of testicular spermatogenic cell distribute uniformly, layer clearly, format normally and there are plenty of sperm in tubule. Interstitial cells appear in interstitial tissue, and whose nucleus are big and round, lie in off-normal. In the model group, seminiferous epithelium exuviate, thin, appear vacuoles, and the cells distribute disorderly. The worse one's a great quantity of spermatogenic cells shed, form multinucleated giant cells and attach in sustentacular cells or shed into the tubule. The blood vessels which in interstitial tissue become more and vessels'wall become thicken. Inflammatory cell infiltrate around vessel and contorted seminiferous tubules. The olive oil group, seminiferous tubule damaged, spermatogenic cell layer reduced and no sperm appeared. The similar situation happen in low-does shugao group and midst-does shugao group. In the high-does shugao group, pathological changes of mouses'testis relieve, and spermatogenic cell shed. To compare with the Sodium Chloride group, spermatogenic epithelium layer reduce, and sperm appear in tubule rarely. The six weeks time, testis and seminiferous tubule are normal in the first group. In the second group, the pathological changes of seminiferous tubule still exist, but better than the three weeks group. Though the spermatogenic cell layer decrease, spermatogonium layer can be seen clearly and interstitial cell increase relately. In the third group pathological changes turn well, there are spermatogonium layerers clearly, and sperm appear in tubule rarely. pathological changes turn well and sperm exist in the forth and fifth groups. Seminiferous tubule distribute uniformly, spermatogenic cell shed rarely, and some sperm exist in the sixth group. The nine weeks time, seminiferous tubule are normal in the first group. In the second group the shed of spermatogenic cell decrease and spermatogenic cell layer increase, and the sperm appear in most of seminiferous tubule, no inflammatory cell infiltrate. In the third group, the forth group and the fifth group, spermatogenic cell layer recover to normal status, spermatogenic cell shed rarely, and sperm exist. Testicular spermatogenic cell distribute uniformly, layer clearly, format normally and there are plenty of sperm in tubule. Most of sperm exist and to show active sperms create. 2. To detect the serum INHB and FSH, it is not different among all of groups. Conclusion: 1.The stability of replicative model is good. 2. the olive oil and shugao play a antag role in the occurrence of experimental autoimmune orchitis ( EAO), but the effection of jiaohao relate with the dosage. High-dose shugao is best. When at the ninth week, it recover basely. |
