Objective:The key reason that makes Chronic hepatitis B difficult to be cured is immune disorders/defects with HBV infected, So the final aim is eliminate HBV and get Immune response. In Chinese medicine, the main mechanism that cure the Chronic hepatitis B is almost througth producing immune recovered. In this study, the influence of TCM decoctions (Liuweiganlu Tang and sijunganlu tang) on the DCs derived from peripheral blood and T cells cultured in vitro in the patients with Chronic hepatitis B infected in phases of immune tolerant and immune clearance. This research is realized by Chinese Pharmacological experiments in plasma, based on the model of DCs and T cells, from which we can detemine a better one between Liuweiganlu Tang and sijunganlu tang to improve the function of immune cell with chronic HBV infection as well as recover Immune response and cure Chronic hepatitis B. Furthermore, this study provides a basis of speculation and research for Chinese medicine to enhance clinical treatment performance on Chronic hepatitis B. Besides, it also supplies certain guidance as well as experimental criterias for a further research and development on new Chinese medicines.Method:To observe the influence of Drug plasma of Chinese Traditional Medicine on the function change of immune cells derived from peripheral blood, PBMCs(peripheral blood mononuclear cell), the source of peripheral blood, was collected from patients with Chronic hepatitis B infected status in different phases to separate DCs and T cells and culture them in vitro in this research. Then Flow cytometry is adopted to detect the expressions of surface cells(HLA-DR,CD80,CD86,CD1a) in DCs, and that of CD28 in T cells. Futhermore, ELISA kit is also employed for IFN-a secretion level detection, which contained in the supernate of DCs.Results:1.The number of the DCs derived from peripheral blood in the patients with HBV-infected status in immune tolerant and immune clearance group is reduced evidently comparing to the control group with healthy persons (P<0.05) when they are cultured in vitro for seven days, and this phenomenon is much worse in the former phase.2. The expression rate of CD80, CD86, together with the capacity of secretory cell factor IFN-α, which are surface cells of DCs, and expression rate of T cells CD28 in patients with HBV infected status in immune tolerant and immune clearance phase are significantly lower than that of the healthy control group (P<0.05).3. With the intervene of blood plasma composed by 10% middle dose Liuweiganlu Tang and 10% small dose sijunganlu tang, part of DCs phenotype and expression rate of T cells CD28 have increased, which proves that both the two therapeutic methods have good effects, and the former one can obtain better performance on improving the expression rate of surface cells in DCs.4. By ELISA detection, it is found that DC secrete more IFN-a when blood plasma is used, but the improvement is not quite obvious (P>0.05).Conclusions:1. The number of the DCs derived from peripheral blood in the patients with HBV-infected status in immune tolerant and immune clearance group is reduced evidently comparing to the control group with healthy persons, and this phenomenon is much worse in the former phase.2. The number and function of DCs and T cells are defective or low in the patients with HBV-infected status in immune tolerant and immune clearance phase, which further testifies that the mechanism of HBV infection immune tolerance and the dysfunction or defect of organism immune cells are closely related.3. Tonify kidney and detoxification as well as strengthening spleen and detoxification have some impact on recovering the function of DCs derived from peripheral blood and T cells in the patients with Chronic hepatitis B infected status in immune tolerant and immune clearance phase.
