AIM:The objective of this study is to investigate the effect of recombinant human interleukin?3(rhIL?3) on the proliferation of megakaryocytie line?IEL~ the interaction of rh[L?3 and rhTB() (recombinant thrombopoitin) and the mechanism of HEL cells?proliferation affected by intcrleukin?3.METHODs: 1. The effects of rhIL?3 on the growth of HEL cells were detected by cell counting oxperimont~ MTT colorimetric assay and methyl green 梡yronin Y cytochemical method. 2. The interaction of?rhlL?3 and rhTPO on the growth of HTTh cells was detected by cell growth curve and MTT colorimotric assay.3. The effect of rhIVl3 on the expression of c梞pl mRNA in IIEL cells was observed by RT梉~CR. RESULTS: 1. Results of?the effect of rhlL?3 on theproliferation of HEL cells: (1). From the growth curve, the numbers and the proliferation rate of I]EL cells in IL?3 group were more than that in control group. In logarithmic growth phase, the doubling time of IL?3 group was shortened in average by 7.5 hours comparing with control group. (2). In experiment group treated with rhIL?3 50, 100, l5Ong/ml, the numbers of HEL cells respectively were 1. 21, 1. 5~, 1. 25 times as many as that in the controlgroup, and the quantities of absorbance respectively were 1. 15, 1. 26, 1. 31 times4as many as that in control group. (3). The result of methyl green梡yronin Y cytochemical method showed that DNA stained as blue (or green) and RNA stained as red. After treated with rhIL?3 lOOng/ml, the integral optical density of DNA in experiment group was 1.38 times as many as that in control group, significantly different (PO. 05).2. Results of interaction of rhlL?3 and rhTBO on the growth of 1111 cells (1). From the growth curve, the numbers and the proliferation rate of IIEL cells in [L?3+TPO group were more than that in IL?3 group and control group. In logarithmic growth phase, the doubling time of IL?3+TB() group was shortened in average by 5 hours versus IL?3 group and 12.5 hours versus control group. (2).
