Background The hepatic portal blood flow including portal vein often to be occluded temporarily during abdomen operation especially hepatobiliary operation. Studies have demonstrated that to occlude portal vein may cause the alimentary canal gore, the bacterium of intestines unbalance, a great deal of endotoxin come into being and gut mucosal barrier damage. Those reasons can lead to intestinal endotoxemia and liver damage. Studies also have demonstrated that octreotide can protect lung damage induced by endotoxin following intestinal obstruction and acute necrotizing pancreatitis in rats. Wheather or not octreotide have protective effect on liver damage induced by intestinal endotoxin? No such report has been seen. In our exeperiment, The change of plasma endotoxin, tumor necrosis factora (TNF-a ), ALL LDII and the hepatic tisses were observed in rats of liver damage induced by intestinal endotoxinObjective To study the Protective effect of octreotide on the liver damage induced by, intestinal endotoxin.Methods Rats model of 憀iver damage induced by intestinal endotoxin was produced by blocking portal vein blood flow. Fourtytwo Sprague-Dawley (SD) rats were divided randomly into seven.~ 6 l~ Jt~ 38~groups (1)Sham-operation group(group S): only opening the abdomen, not blocking portal vein blood flow; (2)Natural saline treatment group A1(group Al):the portal vein was clamped for 30 minutes;(3) Natural saline treatment group A2(group A2):after manipulating like groupAl,the portal vein blood flow was resumed for 60 minutes;(4) Natural saline treatment group A3(group A3): after manipulating as groupAl,the portal vein blood flow was resumed for 120 minutes;(5)Octreotide treament group B I (group B 1):manipulating as group A1;(6) Octreotide treament group B2(group B2): manipulating as group A2;(7) Octreotide treament group B3(group B3): manipulating as group A3. In octreotide treatment groups, octreotide was adminstered subcutaneously at rate of 6ug/kg and intravenous injection at rate of 4ug/kg before operation and repeated half an hour later. The natural saline treatment groups were given the same volume of natural saline. In every group, plasma endotoxin level, serum TNF- ci concentration, ALT and LDH level were measured, a 1cm1 section of hepatic tissue was cuted for lightmicroscope sample immediately before opreation over. The changes of hepatie tissue were observed with light-microscope.Results 1. The level of plasma endotoxin and concentration of serum TNF-u in different groups rats(see tablel).2.The level of ALT, LDH in diffentent groups rats (see table 2).
