Heat Compound Heat Shock Rat Hsp70 Gene Expression Regulation

There are many descriptions of heat syndrome in the Classic of Internal Medicine .Among the famous"Nineteen items of pathogenesis"four items respectively relates to "fire"and "heat"syndromes. The therapeutic principles of the heat syndrome is that "the heat syndrome in nature is treated with cold drugs in nature and the warm syndrome is treated with clearing methods; the therapeutic methods are "curing the syndrome with salty in flavor and cold in nature, adjusted with sweat in flavor and bitter in nature, converged with sour in flavor and diverged with bitter in nature. These statements have an important influence on the offsprings. Physicians of the past have created many fruitful heat-clearing prescriptions(HCP) based on these principles. Now medical research shows that I-ICP have varieties of effects such as fighting against pathogenic microbe, virus, inflammatory, and relieving heat, regulating sympathetic-suprarenal system etc. But there are less reports concerning molecular biology.Heat Shock Protein(HSP) is a hit issue in the medical circles recently. HSPs are prevalently popular in the whole biosphere .When organisms are under irritable condition such as heat shock reaction(HSR),infected by pathogens, or induced by amino acid , growth factor and heavy metal etc, HSPs will be markedly increased ?HSP is one kind of protecting proteins of the body, highly conservative in all species, called "molecular chaperone"related to some essential functions of the cells. All HSPs are induced mainly by the Heat Shock Factors. Two of the most important HSPs refers to HSP7O and HSP84 .The research on HSP7O and HSP7OmRNA are most widely performed thus far.The process of HSR is similar to the heat syndrome and heat梡oison syndrome .Our study is to probe into the effect of HCP whether it regulates the gene of HSPs or not. We choose two recipes as our study objects: one is white tiger decoction(WTD) which is applied to the syndrome of excessive heat in the qi fen, the other is Antidotal Decoction of Coptis(ADC) which is fit for cases with overabundance of heat梩oxin in the tn?warmer . during the experiment we upbuild two kinds of heat syndrome models of animal separately; one is the HSR rat and the other is the poisoning rat caused by endotoxin. We took several methods such as RT桺CR, immunohistochemical methods and morphological observation on opiand eletryThe results show that:l.Morphologically both the two recipes can relieve the injury of the tissues, but the locations and the details are different. 2 Generanily, the expression of the HSP7OmRNA of rats of HSR are higher than of the rats poisoned by endotoxin. For the models of rats of HSR,both increase apparently the expression of the HSP7OmRNA, and WTD抯 effect is greater than ADC抯. 3. For the model of the rats poisoned by endotoxin, the HSP7O mRNA expression of neither WTD抯 nor ADC抯 group has difference from the control group攕..4Conclusion:The effect of Chinese herbal fighting against the disease in gene level, different Chinese herbal affect different gene, the results are not the effect of the only one gene, but in different ways...
