Qingrejiedu Soup Quality Evaluation Method

The method of quality control for Qingrejiedu-decoction was studied in this paper. Two main herbal medicines, Radix Scutellariae and Radix Puerariae were identified with TLC method and the active constituent of each of them,baicalin and puerarin were determined with HPLC method. Baicalin was analyzed with a mixture of acetonitnle-water- tetrahy-drofuran(20:80:2, phosphoric acid mediate pHz=4) as the mobile phase and 280nm detection wavelength was selected. The results showed the calibration curve ~vas linear in the range of 0.2O8~條O.400~ g and correlation coefficient was 0.9998. The average recovery of baicalin was 97.4%,and puerarin was analyzed with acetonitrile-2%glacial acetic acid(8.5:9 1.5 v/v) as the mobile phase and 250nm detection wavelength was selected.The results showed the calibration curve was linear at a range of 0.424?4.24Op. g and correlation coefficient was 0.9998.The average recovery was 101 .3%.The contents of baicalin and puerarin were analyzed at the same time in QRJDA(QjncI~ejjedu..decoetiofl water cooking) and QRJD (Qingrejiedu - decoction water cooking and alcohol precipitating) samples, and the contents of baicalin and puerarin in QR]DA were higher than the QRJD sample.Above method was simple, sensitive and reproducible and provided a qualitative and quantitative basis for the quality assessment of Qingrejiedu-decoction. Extracorporeal bacteriostasis,antibacterial in vivo,anti-inflammation and febrifugal experiment of QRJ7D were studied and the results of extracorporeal bacteriostasis showed that the minimum inhibitoiy concentration for Staphylocccus aureus of QRJD was O.3375g1mL,for Escherichia coil was 0.6750g1mL,and MIC for Staphyiocccus aureus of QRJDA ~ 0.1688g/mL,for Escherichia coil was 0.3375g/mL. The results of antibacterial in vivo showed mortality of the control group was 90%,ASA group was 20.0%, QRJD group was 60.0%, QRiDAgroup was 30.0%,QRJD. QRJD A and ASA groups had a significant imparity compared with the control. QRJD group and QRJDAgroup had no significant imparity in statistics .The results of anti-inflammation showed the inhibition of ASA group was 44.19%,QRJD group was 39.89%, QRJDAgroup was 44.91%, QRJD. QPJDA and ASA groups had a significant imparity compared with the control. QRJD group and QRJDA group had no significant imparity in statistics. The results of the febrifugal showed QRJD QRJIM and ASA groups had a significant imparity compared with the control,and had no significant imparity A between QRJT) group and QR.fl) group.. A new methodology conforming to the theory of traditional Chinese medical and pharmaceutical science,we studied QRiDAand QRJD not only by a qualitative and quantitative test but also by pharmacological experiment and provided basises for the clinical application and the quality assessment of the composing herbs.
