The creation of GanZhiKang Capsule(GC) was guided with the theory of TCM and combined with the achievement of modern pharmacology.lt had benefitical effect on fatty liver. In order to expound its pharmacological action and probe into its mechanism of action,the task had observed the prevention and cure function for GC in the treatment of nonalcoholic fatty liver. It provided scientific basis for clinic and laid a foundation for the further research and exploitation of new drug. Method60 male SD rats were divided into 6 groups at random after feeding for a week, 10 rats made a group.Normal control group(group N) feed common diet,the other five groups are large dose GC group(group G1),middle dose GC group(group G2),small dose GC group(group G3),Dongbaogantai Tablet group(group D) and model group(group M).They were all feed high fat diet .After two weeks.the other five groups were filling with GC at different densities.Group G1, G2. G3 were filling 3g/100g.d, 1.5g/100g.d, 0.75g/100g.d once a day.Group D was filling with DT suspension 0.09g/100g.d,once a day.Group M was filling with normal saline 1 ml/100g.d.All the rats were killed after anesthetized at 12 weeks after experiment.The serum should be separated quickly and liver tissues were obtained.The following were detected in each groups:the serum lipid(TG, TCH, FFA, HDL),liver function(ALT,AST, TP, A).One part of liver was for homogenate to detect MDA, SOD, GSH, VitE, TG.the other part was used as paraffin pathological sections and for electron microscopy.The result of the experiment need T-test and Ridit-analyse. Result1.Compared to the model group,GC group could control the body weight of the modeled rats,reduce liver index and there was no significant difference among GC groups and group N.The reduction of liver index in group GI was obviously better than those in group D(p<0.05).2.ALT, AST in group M increased.There was no significant difference among GC groups and normal control group.The reduction of AST in group GI was obviously better than that in group D(p<0.05).There was no significant difference in TP, A among the 6 groups.3.Compared to the group N,TG, FFA of group M increased.The level of HDL is lower than group N(p<0.01).There was no significant difference among GC groups and group N in TG, TCH, FFA, HDL.There were obviously differences on the reduction of serum TCH and the reinstatement of HDL between group GI and group D(p<0.05,p<0.01).4.In the liver tissues of group M,MDA, TG increased obviously compared to the group N(p<0.01),but the level of SOD, GSH, VitE reduced obviously.The level of TG, MDA, SOD, GSH, VitE in group GI and group 62 were to near normalrange,and there was no significant diffenence with group N.5.In model group,fatty degeneration of liver tissues in different extents was observed by light microscopy.There were a lot of fat bubble,inflammation in lobule of liver and most of the portal area,necrosis in punctate and patchy were also observed.Serious fat drop accumulation was found and swelling of mitochondria,dilatation of endoplasmic reticulum also were observed by eletron microscopy.In GC groups,the reinstatement of ultrastructure of liver cells and reducing of fatty degeneration in liver cells was observed. According to observing pathological sections,the reinstatement of fatty degeneration in group GI is better than those in group D(p<0.05). ConclusionGC has a better antifatty-liver efficacy.The protection of the drug may be associated with:1 .Regulating and improving lipid metabolism,Rationally distributing of fat,which could avoid the form of fatty liver. (l)Reducing TG in serum and liver tissues.Inhibiting the absorption of exogenousTG. Accelerating the excretion of TGReducing the synthesis of TG. (2)Improving the level of HDL.Promoting the reverse transport and metabolismof cholestein.2.Reducing the content of FFA.Improving the function of liver to oxidate fatty acid.Restraining lipid peroxidization.Reinstating the activity and content of SOD > GSH, VitE.Preventing the development of...
