| ObjectiveHepatic fibrosis is the serious pathologic feature of every chronic hepatosis. It can develop to cirrhosis, and it is responsible for liver caner. The method that can be used to therapy it is very limited. We based on clinical practices and relative theories about modern medicine and TCM to observe the effect of anti-fibrosis Capsule FuYuanYuGan on experimental liver fibrosis rats and possible mechanisms. Method1. 70 Wistar rats were divided into six groups randomly ,normal group(group A),model group(group B), colchicine treatment group(group C ),large dose Capsule FuYuanYuGan treatment group(group D),middle dose Capsule FuYuanYuGan treatment group(group E),small dose Capsule FuYuanYuGan treatment group(group F).Group A has 10 rats .Except it ,there were 12 rats in other groups .Except for A ,the every rat of other five groups was modeled by multiple factors(The rats were injected into CCL4 under its skins two times a week and fed on high fat low protein diet).The group A were injected into sweet oil under its skin and fed on normal diet .Until the end of six weeks, the liver fibrosis model were formed. From the second day of modeled, group C were infused with colchicine and Capsule FuYuanYuGan was given in group D (8g/kg.d), group E (4g/kg.d),group F(2g/kg/.d).Group A were treated with 0.9%Nacl.Then all rats were given to drug to collected the blood from hearts to get the value of HA,LM,PIIIP in serum and got the liver tissues to be studies with pathological observation.2. In order to carry out the experimental study on toxicity of Capsule FYYG, 20 mice were given this medicine 24g/kg.d by stomach two times a day. And we took investigations of general condition and dead rate for 7 days.3. To study the long-term toxicity test, 60 rats were divided into three groups randomly, that is normal group, large dose Capsule FYYG group and small dose Capsule FYYG group, and there were 20 rats in every group. The normal group were given normal feed, and the other two groups were treated with Capsule FYYG 8g/kg and 4g/kg differently for 3 months. The general condition was observed, such as mental state, appetite, hair, stool, and moving, etc. At the end of the experiment, all of the rats were killed after the last time when the rats were treated with medicine. And the serum was collected to observe blood picture, function of liver kidney, and other important organs were observed, too. The other half number of the rats was killed in two weeks when the rats were stopped taking medicine. As the investigation of convalescence, the same value as the rats in one group which were killed firstly was observed.ResultsExcept for the normal group, the serum value and HYP of the other groups were raised, but those of treatment groups were significantly lower than model group,were raised, but those of treatment groups were significantly lower than model group, (P<0.01). The value of group D was lower than that of group C, named by HA (P<0.05),LM (P<0.01),PCIII(P<0.01). The level of serum PCIII of group E and F were lower than that of group C. The level of LM of group D was lower than that of group F (P <0.01).Pathological observation : The proliferation degree of collagenous fibers of treatment group was lower than that of the model group, especially that of group D,(P<0.01).Conclusions1 Capsule FuYuanYuGan shows definite protective and therapeutic effect on experimental hepatic fibrosis model rat.2 Capsule FuYuanYuGan had no liver toxication. |
