The Gold Shuiqing Rat Mesangial Cells

Objective Jinshuiqing is a traditional Chinese medicine prescription to treat the glomerular disease. This experiment observe the prescription's poison and its effect on proliferation of glomerular mesangial cells(MC), the level of Interleukin-6(IL-6) and type IVcollagen , the MC apoptosis and the cell cycle of the rats. Thereby it will offer some experimental basis for applying it in clinic better, supplying a kind of new method of traditional Chinese medicine theory basis study for curing this disease.Methods The rat's glomerular mesangial cells were divided into seven groups: normal control, dexamethasone(Dex) contraband the Jinshuiqing groups ranged from ( 1000, 500, 250, 125, 62. 5u g/L ). Then observe the Jinshuiqing's poison with trypan-blue staining tinction and assess the cell proliferation of glomerular mesangial cells by MTT spectrometric assay; Through the fluorescent and ultratructure to observe the morphological variation characteristics of apoptosisbody; to observe cell cycle and the ratio of the apoptosis with the FCM;The level of Interleukin-6(IL-6)and type IVcollagen in rats were assayed by the sandwich Elisa method.Results1 Jinshuiqing has no significant poison to the MC in the experiment at 24hu 48h and 72h.2 The OD results of MTT assay at 24h% 48h show Jinshuiqing can inhibit the proliferation of MC. High dose is same to Dex (P<0. 05),and the low dose does not. Compared with normal control ,there is a remarkable difference(P<0. 05).3 The morphological variation characteristics of apoptosis can be seen through the fluorescent and ultratructure observation.4 Jinshuiqing can inhibit the proliferation of glomerular mesangial cells with delaying the cycle of cells in concentration-dependent manner. It raises the number of cells in Go-d stage,and reduces the number of cells in S and C2 stage.The results present Jinshuiqing can delay the MC from Go-G, stage to S stage.5 - Jinshuiqing can significantly inhibit the level of Interleukin-6(IL-6) and type IVcollagen secreted in glomerular mesangial cells.Conclusion The experiment expounds MC is the important target cell for action of Jinshuiqing . It can inhibit the proliferation and induce the apoptosis of MC.The mechanism of Jinshuiqing maybe relate to it can delay the progress of cell cycle, and prevent the level of Interleukin-6(IL-6) and type IV collagen.The function has concentration-dependence.These findings may provide part of experimental basis for clinic use of Jinshuiqing on the level of cell and molecu.Meanwhile it also presents a way for the combination of TCM and modem scientific knowledge as well as for the development of TCM.
