| ObjectiveAs to treatment of acute focal cerebral ischemic, thrombolysis is publicly taken as one of effective therapies, which has been increasingly regarded, however, reperfusion injury followed it confines the apply clinically. Modern researches indicate that a series of decompensations including excessive generation of free-radical, influx of calcium through voltage-gated calcium channel, triggering the release of neurotransmitters, inflammation response and programmed cell death are the primary pathogenesis of ischemic-reperfusion damage and lead to the death of neuron.. The ischemic-reperfusion damage is a multi-level pathologic and physiological process. Chinese traditional medicine(TCM) compound has various chemical component and broad biology activity, which provides the possibility on theory and practice for holding up the multi-approach and multi-level injury from ischemic-reperfusion damage. Ditan decoction is one of the head Chinese traditional medicine compound which can treat the brain diseases. It has been indicated by a few of studies clinically that the Ditan improvement decoction is effective in treating cerebral ischemic, therefore the study investigates the effect and explore the protective mechanism of Ditan decoction, which can provide scientific proof to enlarge the clinical apply of the same kind Chinese traditional medicine and new preparation of cerebral protection. MethodsFifth-four of male Wistar rats weighed 250~300g(Provided by Medical College in Wuhan University)were randomly divided into three groups: sham-operated group, control group and treatment group. The focal cerebral ischemic-reperfusion model was established with thread embolism of middle cerebral artery. After suffering 2 hours occlusion and following 70h reperfusion ,we observed the protect effect of the Ditan decoction by the change of the whole situation, infarct volume, cerebral water content, damage degree of brain function, the pathology of brain tissue with light microscope., apoptosis with Flow Cytometry, the expression of TNF- a (tumor necrosis factor-α) with immunohistochemitry techniques. Results1.The decoction can significantly improve the whole situation, brain function of focal cerebral ischemic reperfusion. After operation, the rats in sham-operated group is good in psychosis and hair cluster. Compared with the sham-operated, the control group and treatment group were less poor in energy, dimmer in hair, reductive in spontaneous activities and bite , and sup, degressive sensitivity on environment, however, the whole situation above of Ditan decoction group were gradually improved.2.At the 72h after operation, there was no neural function impair in thesham-operation.The neural function score in the treatment group was markedly lower than that in the control group (P<0.05)3. At the 72h after operation ,as to the rats of focal ischemic-reperfusion, compared with the control group, the Ditan decoction can lighten their cerebral edema and cut down their infarct volume. There was thoroughly distinct difference between the two groups(P<0.01)4. At the 72h after operation,there were a spot of expression of TNF-α in the sham-operated group with immunohistochemitry techniques. The expression of TNF-α in the Ditan decoction group is less marked than that in the control group(P<0.05).5. At the 72h after operation,the incidence of neuronal apoptosis measured with Flow Cytometry in the control group and the treatment group had remarkably more increasing than that in the sham-operated group (P<0.01) . The Ditan decoction group had less decreasing than the control group. There were quietly marked difference at the time of 72h between the two groups (P<0.05) .All above suggest Ditan decoction can restrain apoptosis.Conclusions1.The changes of the whole situation, infarct volume, cerebral water content, damage degree of brain function and the pathology of brain tissue with light microscope indicate that Ditan decoction has the brain protection effect for cerebral ischemic-reperfusion damage.2.By the mechanism... |
