Objective: The experiment aims to find out the physiological role of aquaporin-2 in the body water metabolism through the study of aquaporin-2' s expression in the renal collecting duct principle cells and further provides the experimental evidence of occurrence mechanism of kidney deficiency polyuria.Method: Through the method of destroying hypothalamus of paraventricular nuclei and superventricular nuclei to creat the rat model of kidney deficiency.Using the method of collecting 24 hours urina to measure the urinary quantity and osmotality;Using the mothod of radioimmunossay to measure plasma VP level; Using the method of HE staining to observe the state of kidney collecting' duct; Using the mothod of immunohistochemistry to detect the protein expression quantity of aquaporin-2 in collecting duct principal cell; Using the method of ONE-STEP PCR to detect the mRNA expression quantity of aquaporin-2 in the kidney. Compare the change states of kidney deficiency graup to the control group.Result: Compared to the control group, the rats of kidney deficiency had the evident symptom of diuresis(P<0.01) , the lower urinary osmotalily(P<0.05) and plasma VP level (P<0. 05). HE staining of collecting duct under the light. microscope displayed: collecting duct shows no change; the mRNA and protein quantity of aquaporin-2 in collecting duct principal cells of kidney deficiency rats were less than that of the control group(P<0.05) Conclusion:1. Kidney deficiency can increase urinay quanlity and decrease osmotality2. Kidney deficiency can decrease significantly the expression of mRNA of aquaporin-2 to in renal collecting duct.3. Kidney deficiency can decrease significantly the expression of protein of aquaporin-2 in renal collecting duct.So, we can conclude from above that Aquaporin-2 may have important physiological role in t he water reabsorption and reduction of Aquaporin-2 may be one of the pathological mechanism of kidney deficiency polyuria.
