Objective:In this experiment,from the perspective of aquaporin,Liuwei Dihuang Wan was used as a positive control group to explore the effect of different doses of tortoise-shell glue on rats with kidney yin deficiency pathology model,aiming to enrich the physiopathology of kidney yin deficiency syndrome and to further To clarify the pharmacological action of tortoise shell gum on kidney-yin deficiency syndrome provides a certain theoretical basis and some experimental basis.Methods:In this experiment,60 SD rats with a body weight of 200 g were randomly divided into a normal group,a model control group,an experim-ental group(low,medium,and high-dose tortoiseshell gel group)and a positive control group(Liuwei Dihuang Wan group),each group 10 Only,using thyroxine tablets as the modeling drug to prepare a rat model of kid-ney yin deficiency,using the method of adminis-tration while modeling,thyroxine tablets in the morning,tortoise shell gum or Liuweidihuang pills in the afternoon,continuous administration for 21 days.During the experi-ment,observe the general signs of rats,including the body weight,mental state,coat color and gloss of rats in each group before and after administr-ation;record the 24-hour water intake,food intake and urine output of rats in each group after the last administration to detect large Anal temperature of rats;after the experiment,the content of AQP1,AQP2,AVP,c AMP,c GMP in the serum of each group of rats was detected by Elisa method,the pathological changes of kidney tissue of each group were detected by HE staining method,and the immunohistochemical method was used.The expre-ssion of AQP1 and AQP2 protein in the kidney tissue of each group of rats.The expression of AQP1 m RNA and AQP2 m RNA in the kidney tissue of each group of rats was detected by fluorescence quantitative PCR.Results:1.General signs: tortoise shell glue can improve the general signs of rats.The rats in the tortoise shell glue dosage group had good mental condition,shiny hair,no vertical hair,no back bowing and no fighting behavior.The weight of rats increased(p < 0.01),anal temperature and24-hour drinking water decreased,urine increased(p < 0.01),and food intake decreased(p < 0.05).2.the content of c AMP and c GMP in rat serum: the content of c AMP and c AMP/c GMP ratio in tortoise plastron medium,high dose group and Liuwei Dihuang pill group are lower than that in model group(p < 0.01),while the content of c GMP in tortoise plastron medium,high dose group and Liuwei Dihuang pill group is higher than that in model group(p < 0.01).3.levels of AQP1 and AQP2: compared with the model group,the levels of AQP2 and AVP in serum of rats in the high dose group of tortoise plastron glue and the group of Liuwei Dihuang pills were significantly decreased(p < 0.01),and the content of AQP1 was Elisa decreased(p <0.05).Immunohistochemical examination showed that the expressions of AQP1 and AQP2 in renal tissue of rats in the tortoise shell glue dosage group decreased after tortoise shell glue intervention compared with the model group.Compared with the model group,the expression of AQP1 m RNA and AQP2 m RNA in renal tissue of rats in the tortoise plastron dosage group was decreased after tortoise plastron intervention by fluorescence quantitative PCR.4.HE staining results of rat kidney tissue: Compared with the normal group,the model group has glomerular hyperemia,enlarged glomerular sac,metabolic dilatation of renal tubules and infiltration of adipose tissue.Compared with the model group,the glomerular hyperemia in the low-dose group of tortoise shell glue is slightly relieved,the capsule cavity is slightly enlarged,the renal tubules are metabolically enlarged,a small amount of adipose tissue infiltration can be seen,the glomerulus in the medium-dose group of tortoise shell glue,the high-dose group of tortoise shell glue and the Liuwei Dihuang pill group are slightly hyperemia,the capsule cavity is not obviously enlarged,and the renal tubules are not obviously expanded.Conclusion: The pathological model of kidney yin deficiency can be successfully prepared by intragastric administration of thyroxine tablets to rats.Carapax et Plastrum Testudinis can correct the changes of basic physical signs and the contents of cyclic nucleotides and the expressions of AQP1 and AQP2 in rats with kidney yin deficiency.High dose of Carapax et Plastrum Testudinis has the best effect.
