Arthrosclerosis is the pathologic foundation of much cardiovascular disease. In recent years, the research of serum pharmacology on Smilax glabra Roxb.(SGR) and Rhaponticum uniflorum (L.) DC. shows that both of them have the effect of anti-atherosclerosis, but we don't know what component in the serum sample. So our study is based on the research of serum pharmacology of SGR and Rhaponticum uniflorum (L.) DC., adopt separately Beagle and SD rat to carry on the preliminary study of on serum pharmacochemistry of SGR and Rhaponticum uniflorum (L.) DC.. OBJECT 1 Study preliminarily on serum pharmacochemistry of SGR. 2 Study preliminarily on serum pharmacochemistry of Rhaponticum uniflorum (L.) DC.. METHOD 1 The study on serum pharmacochemistry of SGR using beagle as experimental animal Use Beagle as experimental animals: take control blood from vein of beagle's forelimb before administering drug, take blood containing effective constituent from SGR on different time after administering drug. Detect all of the samples by high performance liquid chromatography (HPLC). Compare he chromatograms of control serum sample and serum samples containing effective constituent from SGR that get on different time after administering SGR, in the course of experiment, we improve the quantity of the drug, the method to deal with the serum samples and the condition of HPLC. 2 The study on serum pharmacochemistry of SGR using SD rats as experimental animal Use SD rat as experimental animals instead: the experimental animals are divided into control group and the group of administering drug. Serum samples are analyzed by HPLC. We get the spectrogram and chromatogram. Compare the chromatogram of control serum samples with other samples. 3 The study on serum pharmacochemistry of Rhaponticum uniflorum (L.) DC. using SD rats as experimental animal We adopt the SD rats as the experimental animals. Animals are divided into control group and the group of administering drug. The SD rats of administering group are per so extract of Rhaponticum uniflorum (L.) DC. three times. Serum samples are dealt with perchloric acid. Samples are analyzed by HPLC. Compare the chromatogram of control serum samples with other samples. In the course of experiment, we improve the condition of HPLC, the method of dealing with the serum samples. The sample of extract of Rhaponticum uniflorum (L.) DC. is detected by liquid chromatography-mass spectrograph (LC-MS). RESULT 1 After studying the chromatograms of the samples from beagle, we find there is no obvious difference between control serum sample and serum samples containing effective constituent from SGR that get on different time after administering drug. 2 We get the samples from SD rats. Comparing with the chromatograms of control serum sample, we find characteristic chromatographic peak at retention time about 14.7min on the chromatogram of serum samples containing effective constituent from SGR. Compare with the chromatograms of serum samples containing effective constituent from SGR, we find corresponding chromatographic peak at retention time about 14.7min on the chromatogram of control serum sample adding SGR. UV spectrum shows that both of the chromatographic peaks have the absorption maximum at about 260nm. 3 We get the samples from SD rats. After improving the method, comparing with the chromatograms of control serum sample, we find characteristic chromatographic peak at retention time about 14.6min on the chromatogram of serum samples containing effective constituent from SGR. UV spectrum of this characteristic chromatographic peak shows that the absorption maximum at about 260nm. 4 We get the samples from SD rats. After improving the condition of HPLC, comparing with the chromatograms of control serum sample, we find characteristic chromatographic peak at retention time about 20.7min on the chromatogram of serum samples containing effective constituent from SGR.
