Purpose: To investigate the expression of intercellular adhesion moleule-1(ICAM-1)after retinal ischemia reperfusion in rabbit and the injury of retinal formation and function. Methods: Select 50 two months old healthy rabbit without disease of the eyes,which are divided into ten groups with random ,that is the normal control group,the perfusion group without high pressure, ischemia reperfusion 0 ,3 ,6 ,12, 24 ,48 hours, 4 and 7 days group.Every group have five rabbits(ten eyes).Ratinal ischemia was induced by increasing the intraocular pressure (IOP).After madding rabbit models, animals were examined by F-ERG, subsequently kill the rabbit and get the eyes at once, and made the sample HE staining and immunohistochemical (SABC)staining .Select the rabbit of normal group,IRI 3hours and 7 days group to do electron microscope examination. The result of SABC staining were analyzed by compute picture analytic system. Result: 1.F-ERG:There was no difference between the group A and B ,the b-wave almost vanished at IRI 0 hour and increase partially at 3 hours, after that b-wave fall down persistly and progressively. 2.Light microscope: there was edema in ganglion cell layer, and ganglion cell decrease progressively. A few of neutrophils infiltrated in retina at 6 hoursafter reperfusion. As time passing, the number of infiltrated neutrophils increased, and a lot of neutrophils were observed at 24 and 48 hours, then decreased progressively. 3.Electron microscope:After ische reperfusion,the ganglion cells of retina show that the mitochondrion was vacuocated and membranous discof outer segment was damaged.The longer had ischemia continued ,the more serious was the reperfusion injury. 4.The expression of ICAM-1:The minute expression of ICAM-1 could be seen in the A and B group ,ICAM-1 expression began to increase after retinal ischemia for 60 minutes,peaked at 24 hours after reperfusion,and decreased thereafter. Conclusion: ICAM-1 expression in retina upregulated,followed with the increasing of infiltrated neutrophils in the early stage of retinal ischemia reperfusion , which contributed to retional ischemia reperfusion injury.
