Aerial Parts Of Gentiana Chemical Constituents Of Bear Bile Powder, Separation And Determination Of Tauroursodeoxycholic Cholate And Taurocholate Goose Sodium Deoxycholate

1 Qinjiao is the roots of Gentiana macrophylla Pall., Gentiana straminea Maxim.,Gentiana crassicaulis Duthie ex Burk and Gentiana dahurica Fisch. Which belongs to Genus Gentiana. It is an important traditional Chinese drug . The main component gentiopicroside have the effects of anti - inflammatory, stopping pains, protecting liver and Lidan. And its prospects is broad. In order to make full use of the resource and expand the using parts of Qinjiao ,we carried out a systematic research on the chemical constituents in the aerial parts of it .Six compounds were achieved by using several separation methods. The structures of them were authenticated by the means of spectroscopy (UV,IR,MS,~13C-NMR,~1H-NMR, HMBC,HMQC etc.) The optimum extract method was selected by using of orthogonal experiment ,and the optimum model of resin was selected by comparing the adsorptional ability of different resins to gentiopicroside. Meanwhile by doing adsorption and unadsorption experiments, the best technological conditions was determined. Besides the content of gentiopicroside in different aerial parts was determined by HPLC. We have done some basic researches for the important project of Shannxi educational department -"the comprehensive development and utilization of the aerial part of Qinjiao".2 bear biliary drainage powder was the dry bile of Vrsus arctor L or Selenarctor thibetanrs Cuvier. It is used as bear bile in clinic, and belongs to the first category new medicine which was ratified by the Ministry of Health. The main components were combine cholic acid. We get the sodium tauroursodeoxycholate and sodium taurocheno- deoxycholate from bear biliary drainage powder by using of the classical separation method and high performance preparative liquid chromatography. Meanwhile we determined the content of sodium tauroursodeoxycholate and sodium taurochenodeoxy- cholate in bear biliary drainage powder by ELSD-HPLC .
