| Rheum officinale Baill , belonging to the Polygonaceae ,is one of the commonly used Chinese traditional medicines as cathartic, hemostaticand astringent. Its main component consists of anthraquinones,tannins flavo- noids and pyrones etc.Recently activity anti-virus of Rheum officinale Baill was found to provide a convenience and scientific guide to treat the various diseases caused by virus. Focusing on chemical structure and the method of extracting and purifing its activity anti-virus ,we carried out following studies.Dry plant powder was extracted with EtOH at room temperature. The solution was concentrated under reduced pressure to afford EtOH extract.The EtOH was extracted with CHCl3,AcOEt and H2O;AcOEt fraction subjected to column chromatographer on the sinical gel.Using CHCl3:MeOH as eluent,several compounds were separated and five among them were identified as follows: I : (-)-epicatechin-3- O- gallate ; II : (-)- epigallocatechin-3- O- gallate); III chrysophanol; IV:emodin:V:aloe-emodin. Especially,compound I and II show great effect of anti-virus.But because of comparability of their chemical structure and property ,it is very difficult to separate them. After a lot of experiments we succeeded to get compound I and II.Furthermore ,content of compound II in extract was determined through HPLC.Chromatographic column:Alltech C18 (250mm×4.6mm , 5μm) ; mobile phase:water:methanol:HAc(98:1:1);flow rate 1.0ml·min-1 Colum temperat-ure:30℃;Wavelength:280nm.The result indicated that the method is easier and convenient,accurate and reliable,which can effectively separate compound I and II .These experiments provided the data for study on medicine anti-virus in the future.Through th improvement of the modern technique,the traditional Chinese medicine becomes more effective and convenient .To make the traditional adapt to the demand of society is the aim of our study. |
