Objectives: To study the effect on the contents of alkaloids in the decocting process of RALP[WPRA, BPRA, CRAP], approaching possibility that MA takes material base on the warming yang efficacy of RALPMethods: In the direction of clinical application of RALP, studying RALP [WPRA, BPRA, CRAP], applying analytical technique of UV and HPLC and SPSS software to handle data, selecting the contents and dissolution rate of total alkaloids-ester diterpenoid alkaloids-DDA indexes to confirm controlling indexes of decocting conditions, the best DT of stable active component. Studying the effect of solution circumstance of decoction in RALP on hydrolyzable rules of MA by the procedure of single factor-compatibility factors-combination factors, approaching the effect of the compatibility of RALP- Radix et rhizoma rhei and RALP-Rhizoma zingiberis on the content of MA. Results:1.Different decocting conditions [water volume, DT.etc. ] have comparatively large influence on total alkaloids and DDA, but no evident regularity.2.DT has significantly effect on the contents and dissolution rate of total alkaloids-ester diterpenoid alkaloids-DDA. The best DT of RALPis 4-6 hours. But at this time the content of MA in the CRAP is higher; and in the BPRA and WPRA ,the differences of both are different contents in the ester alkaloids content and aconitine 0.25-1 hour also quite is stable.3.The MA in the decocting process of different medical materials is the most stable and has comparatively high content, its dissolution rate and content curve was similar to the addition of MA, aconitine and hypaconitine, which had representative in single medical material4.0n the same decocting conditions of RALP, Acid(pH=4.5) inhibits the hydrolysis speed of MA, which is an important characteristics in BPRA and WPRA other than the decocting solution of RALP. Glucose, starch and inorganic salt accelerated the hydrolyse of MA, hydrolyzable speed is Na2SO4>starch>glucose, the hydrolysate of MA might be changed at alkalescent(pH=9.0) ,Rhein and ammonium glycyrrhizate together. AB solution is obtained by simulating the combination conditions of decocting circumstance of RALP, which can remarkably inhibits hydrolyzable speed of MA.5.The compatibility of RALP and Rhizoma zingiberis markedly decreased the content of MA, the ratio of 2: 1 and 1: 2of RALPversus Rhizoma zingiberis decreased the content in MA were less than others ratios. At the same medical material, the identical variety blends the proportion difference to be big .The content of MA is comparatively high and small decreasing range after the compatibility of RALP and Radix et rhizoma rhei, at the same medical material, with the increase in Radix et rhizoma rhei, the decreasing range become large. Conclusions:Having established the controlling indexes of effective factors of decoction under the laboratory conditions, suggesting refer to execute in clinical application.The best DT of stable active component of RALPin clinical application is 4-6 hours, But at this time the content of MA in the CRAP is higher; in the BPRA and WPRA, both of them can...
