Objective: In this paper, we study for the mechanism of traditional Chinese medicine to knee osteoarthritis. We put forward the theory of "Bushen Tongluo" to therapy OA based on the traditional Chinese medicine; We also study the machanism of the protection of Gubikang Prescription to chondrocytes induced by IL-1α. At the same time, we study the therapeutic effect of Gubikang Prescription on knee osteoarthritis.Methods: Cartilages from 6 cases after resection were digested by 0.2% collagenase Ⅱand cultivated in 10% DMEM. The number of chondrocytes gained from wet cartilage, viability, multiplication and morphologic changes were observed. Chondrocytes were divided into different groups, GBK was added to subculture cells and its influence on chondroncytes proliferation in different time was examined by MTT method, and the influence on collagen type Ⅱ mRNA expression were examined by RT—PCR method. The changes of protein levels of collagen type Ⅱ was observed by immunohistochemistry.Sixty cases of knee OA patients who were determined deficiency of kidney and cold coagulation, stagnation of blood stasis were divided randomly into the treatment group and the control group, the former was treated with GBK and the latter with Glucosamine capsule. Four weeks is a course of treament.
