Objective:This study in order to discussion on the effect of osteopontin (OPN) on mRNA expression of aggrecan and collagen type Ⅱ in osteoarthritic Chondrocytes of human knee in vitro, which can further explore the pathogenesis of Osteoarthris,Thus, it can provide a new theory which will be a safer and more efficient approach as a treatment targets for osteoarthritis.Methods:Primary cultures osteoarthritic chondrocytes.The first generation of osteoarthritic chondrocytes of human knee was divided into control group(group A) and experimental group (group B and group C), group B cultured with OPN (100ng/ml)for48h, group C cultured with OPN (1μg/ml)for48h, group A cultured without OPN,Furthermore, we evaluated the effect of OPN on OA chondrocytes, the mRNA expression of aggrecan and collagen type Ⅱ was evaluated by RealTime-PCR. Results:1.Compared to group A, mRNA expression of aggrecan of group B (2.819±1.175folds) elevated,and the difference showed statistically significant (P<0.05)2.Compared to group A, mRNA expression of aggrecan of group C (3.731±0.607folds) elevated,and the difference showed statistically significant (P<0.05)3.The difference of mRNA expression of aggrecan between group B and group C showed statistically significant (P<0.05) Further more, mRNA expression of proteoglycan of group C were higher than which of group B.4.The mRNA expression of aggrecan in osteoarthritic chondrocytes of human knee in vitro were positively strongly correlated with the concentration of OPN interferenced (r=0.751P<0.01)5. Compared to group A, mRNA expression of collagen type Ⅱ of group B (2.886±0.658folds) elevated,and the difference showed statistically significant (P<0.05)6. Compared to group A, mRNA expression of collagen type Ⅱ of group C (4.725±2.895folds) elevated,and the difference showed statistically significant (P<0.05)7.The difference of mRNA expression of collagen type Ⅱ between group B and group C showed statistically significant (P<0.05),Further more, mRNA expression of proteoglycan of group C were higher than which of group B.8. The mRNA expression of collagen type Ⅱ in osteoarthritic chondrocytes of human knee in vitro were positively strongly correlated with the concentration of OPN interferenced (r=0.6761P<0.01)Conclusion:1. OPN up-regulate mRNA expression of aggrecan in osteoarthritic chondrocytes of human knee in vitro. The mRNA expression of aggrecan in osteoarthritic chondrocytes of human knee in vitro was positively strongly correlated with the concentration of OPN interferenced.2. OPN up-regulate mRNA expression of collagen type Ⅱ in osteoarthritic chondrocytes of human knee in vitro.The mRNA expression of collagen type Ⅱ in osteoarthritic chondrocytes of human knee in vitro was positively strongly correlated with the concentration of OPN interferenced.
