| Stroke is a common disease in the old people, and has the characteristics of high morbidity, high relapse and high incidence of causing disability. Furthermore, it may threaten people's lives. The morbidity of acute ischemia cerebral vascular diseases is much higher.The key treatment during acute cerebral ischemic attack is to renew the blood stream, however, ischemical reperfusion injury going with the reperfusion. In recent years, many researches have proved that Chinese medicine can make great progess in prevention and cure of cerebrovascular disease, and what's more, show the great superiority in ischemical reperfusion injury.This research consists of two parts. The former is for Chinese Medical theory research, we give a discussion for the academic origin and achievements of Synopsis of Prescriptions of the Golden Chamber's stroke chapter. We justificate that the theory of this chapter comes of Internal Classic of Yellow Emperor, especially, using the Prescriptions of"houshihensan"which treating for ischemia cerebral vascular diseases and"fengyintang"which treating for acute hemorrhage cerebral vascular diseases as the representation for treating strok breaks a new way for the theropy for strok, also makes a great contribution for the academic development.The later is for experimental study, we chose TGFβ-1 and Caspase-3 as objective, using immunohistochemical technique to observe the expression of TGFβ-1 and Caspase-3 in nerve cell after ischemia and reperfusion. At the same time, we observed Chinese medicine's effect on the expression of TGFβ-1 and Caspase-3.The main results displayed were as follows:1. Observing the injury and necrosis of neuron in the damaged brain tissue by hematoxylin-eosin staining (HE) shows that"Dange"Infusion has protection for cerebral ischemia-reperfusion in mouse.2. The results showed that expression of Caspase-3 were observed markedly in CA1, CA3 and cerebral cortex and the neurons were in brown. Caspase-3 positive cell number and color in"Dange"Infusion group were higher than that in cerebral ischemic reperfusion group. It indicated that"Dange"Infusion can increase the expression of Caspase-3. The effects of"Dange"Infusion on the morphology changes occurred better than model groups.3. The results showed that expression of TGFβ-1 were observed markedly in CA1, CA3 and cerebral cortex and the neurons were in brown. TGFβ-1 positive cell number and color in"Dange"Infusion group were lower than that in cerebral ischemic reperfusion group. It indicated that"Dange"Infusion can decrease the expression of TGFβ- 1.In summary, the above results suggested that"Dange"Infusion has protection for cerebral ischemia-reperfusion in mouse. The mechanism of action is that"Dange"Infusion can enhance the expression of TGFβ- 1 protein and decrease the expression of Caspase-3, which proves that"Dange"Infusion has the neuro-protective effect against ischemia reperfusion injury. |
