Limited by the traditional ideas which herbs can only be used in chronic diseases, traditional Chinese medicine (TCM) is less used in acute asthma.It will be of great importance to improve the effect of TCM on treating acute asthma and understand the mechanism of treating asthma of classical prescriptions. Traditional Chinese materia medica and medicinal decoctions have multiple targets so that they can exert influences on multiple pathological processes of asthma and produce curative effects. With the help of modern biomedical sciences , we can demonstrate the therapeutic effects of Chinese materia medica and elucidate the underlying mechanisms of drug actions. These tudies help us find new drugs for asthma from Chinese materia medica and medicinal decoctions.Shuanglong granule (SL) is prepared on the basis of clinc experience. The SL takes into consideration of mainShuang long (SL)capsule is Chinese drugs preparation based upon an effective medical prescription containing of Ephedra ( Herba Ephedra ) , Chinese Magnoliavine Fruit ( Fructus Schisandrae chinensis ) and Earth worm (Allolobophora Caliginosa trapezoids) ect. The studies include literature review, demonstration of prescription, pharmacology research and mechanism exploration.1. ObjectiveDemonstrate the antitussive, apophlegmatic and anti-inflammatory actions of SL. Observe the anti-asthmatic effects of SL and the relaxation effects of SL on guinea pig tracheal smooth muscles ex vivo.2. MethodsThe influence of SL on cough latent period and cough rate was investigated using strong aqua ammonia-induced cough model in mice to demonstrate SL's antitussive action. The effects of SL on the phenolsulfonphthalein excretion of mice trachea were observed to decide SL's apophlegmatic action. The anti-inflammatory effects of SL were detected by the degree of ear swelling using xylene-induced ear swelling in mice. The anti-asthmatic effects of SL were observed using OVA-induced asthmatic guinea pig model. In vitro experiment,guinea pig trachea strips were prepared. The bronchodilator effects of SL on isolated guinea pig trachea strips with or without the pre-contraction function were studied.3. Results3.1 SL's antiasthmatic effectsTo stimulate with OVA in first day ,SL and Dexamethason Acetate (Dex) could prolong the latent period of asthma induced by OVA. SL at doses of 6.30g/kg and 3.15g/kg was significantly different from the control group(p<0.01).In 6th day,SL at doses of 6.30g/kg,3.15g/kg and the positive control (Dex) were significantly different from the control group(p<0.05). SL has antiasthmatic effects.3.2 Antitussive, apophlegmatic and anti-inflammatory activities of SL3.2.1 Effects of SL on strong aqua ammonia-induced cough in miceSL and Pentoxyverine Citrate could decrease the frequency of cough within 2 minutes caused by ammonia water. The positive control (Pentoxyverine Citrate)were significantly different from the blank(p<0.01). SL at doses of 6.30g/kg,3.15g/kg and TCM positive control were significantly different from the blank(p<0.05).SL and Pentoxyverine Citrate could prolong cough latent period caused by strong aqua ammonia in mice. The positive control (Pentoxyverine Citrate)were significantly different from the blank(p<0.01). SL at doses of 6.30g/kg,3.15g/kg 1.58g/kg and TCM positive control were significantly different from the blank(p<0.05).SL has demonstrated antitussive effects.3.2.2 Effects of SL on SL excretion of phenolsulfonphthalein in mice tracheaSL and the control group could significantly increase the excretion of phenolsulfonphthalein in mice trachea. The positive control group were significantly different from the blank(p<0.01). SL at doses of 6.30g/kg,3.15g/kg 1.58g/kg and TCM positive control were significantly different from the blank(p<0.05).SL could promote expectoration.3.2.3 Effects of SL on xylene induced ear swelling in miceSL could inhibit the degree of ears swelling . The positive control group were significantly different from the blank(p<0.01).SL at doses of 6.30g/kg, 3.15g/kg and TCM positive control were significantly different from the blank(p<0.05). SL have anti-inflammatory effects on xylene-induced ear swelling in mice.3.3 Bronchodilator effects of SL on isolated guinea pig trachea strip SL could antagonize the contraction of ex vivo guinea pig tracheal strips induced by His and Mch. SL has broncho-relaxing effects on resting isolated trachea strips from normal and asthmatic guinea pigs.4. Conclusion4.1 SL's antiasthmatic effectsSL could prolong the latent period of asthma guinea pig induced by OVA.The same time SL could cut down the death rate of guinea pig during experiment time. SL has antiasthmatic effects.4.2 Antitussive, apophlegmatic and anti-inflammatory activities of SLSL shows excellent antitussive and apophlegmatic actions and has anti-inflammatoryeffects on xylene induced ear swelling in mice.4.3 Bronchodilator effects of SL on isolated guinea pig trachea stripSL has antiasthmatic effects. In vitro experiment,SL can antagonize the contraction of ex vivo guinea pig tracheal strips induced by His and Mch. SL has relaxing effects on resting isolated trachea strips from normal and asthmatic guinea pigs.
