Panax ginseng has long pharmacological history in our country. It has a lot of physiological activity. The ginseng is the precious traditional Chinese medicine.It is in the main status among many Chinese patent medicines . Ginsengoside is its main ingredient. HPLC is used to control its quality. Because the content of Ginsengoside is few and easy to be disturbed by other ingredients in the prescription .Therefore the content determination method encounters the big difficulty. The key factor of HPLC method lies in the establishment of chromatograph condition and the preparation of sample solution.The chromatograph condition is mature, but sample solution preparation is difficult. The pharmacopoeia in 2005 version gathers over 20 kinds of traditional Chinese medicine which contain the ginseng,Only two kinds have the method of determination. Its difficulty also lies in the sample solution preparation.Therefore ,the subject's aim is to seek one general sample solution preparation method and give a extensive application .This subject has included several aspects : First part: the research has be conducted to establish a general method of determining ginsengoside Rg₁ and Ginsengoside Re by taking the guilingji capsule experiment as anexample .The general method is : extraction by methynol—the evaporation ofextracts—dissolving of water—lavation of the methylene chloride—extraction of normal butyl alcohol—lavation of 1% sodium hydroxide—Laundering to neutrality by water—evaporation of the normal butyl alcoholextracts—Methynol dissolution—sample solution .Second part: the generalmethod of sample solution preparation is promoted to shenmai injection and yiqi fumai keli. The repeatability, precision and accuracy meets the requirements. Third part: Study on the extraction of Ginsengoside from ginseng with Ultrasonic wave .This method which established in this article could be used as a reference to preparate the sample solution of many Chinese patent medicines including ginsen.
