Objective: To investigate the effects and significance of endothelin(ET),nitric oxide(NO)on hepatic injury of rats with obstructive jaundice. To evaluate the protecive effects on hepatic function and mechanism of Ginkgo bilboa injection (GBI) in rats with obstructive jaundice (OJ ).Methods: Twenty rats with ligated bile duct were at random divided into Obstructive jaundice group(OJ). and Ginkgo bilboa injection group(GBI). The rats were administered 0.75m1/kg saline and 0.75m1/kg Ginkgo bilboa injection since the second day after bile duct ligation.Ten rats had sham-operation. The levels of ET and NO in blood and liver tissue were determined with radioim-munoassay. and colorimetry respectly. Meanwhile,the levels of ALT and TBi in blood were assayed on14day after the bile duct ligation and hepatic histological changes were observed with light microscopy.Results: The levels of ET in blood and liver tissue in OJ group and GBI group increased significantly, compared with SO group . The level of ET in Ginkgo bilboa injection group were lower than those in OJgroup.The levels of NO in blood and liver tissue in OJ group and GBI group increased significantly, compared with SO group . The level of NO in Ginkgo bilboa injection group were lower than those in OJ group. The levels of ALT and TBi in blood serum changed significantly according to the level changes of ET and NO. Histological examination showed that liver cells were damaged remarkably in OJ group. Ginkgo bilboa injection ameliorates the pathological changes of the hepatic tissue.Conclusion: (1) The level of ET and NO in blood and liver tissue increased significantly after the bile duct ligation, compared with SO group. In rat model of experimental obstructive jaundice, hepatic morphology and liver biochemical tests altered significantly (2) From the treatmentof Ginkgo bilboa injection, the levels of ET and NO in blood and liver tissue decreased significantly, liver function and liver cell damage abate.Ginkgo bilboa injection improves liver function and decreases hepatic histopathology induced by OJ in rats. The study showed that Ginkgo bilboa injection is effective in preventing liver damage in obstructive jaundice.
