Jinlianhua(Flos Trollii) ,derived from the dried flower of Trollius chinensis Bunge. and Trollius asiaticus L.,is a commonly used medicine for removing heat and toxic substance. Its effect of therapy was found by monk in Wutai mountain. It is one of the important medicine resources in Shanxi province.Although it is certain that Flos Trollii is a good antibacterial medicine, there are no studies on effective substances and quality standard of Shanxi's Flos Trollii.Using the content of total flavonoids and extraction yield of Flos Trollii as index, the experiment studied on the quality control of Flos Trollii, and a perfect quality standard for Flos Trollii was established. Based on the traditional experience and modern science researches,we decided the most optimum parameters by Genetic Algorithm (GA) in Flos Trollii and established a quality standard for obtaining the valid part. In addition, we studied the pharmacological action preliminarily.The main contents of the papers are as follows:The study of the quality standard in Flos Trollii. Firstly, the experiment identificated the original plant of Flos Trollii by Plant taxonomy ,and the scientific name of the original plant in Lishan was Trollius chinensis Bunge. Secondely, by Macroscopical identification , Microscopical identification and Physical and Chemical identification, we found the characters of Flos Trollii. Moreover, the content of total flavonoids was determined by visible spectrophotometry. The content of total flavonoids was 8.59%.A perfect quality standard to assure the effects and satety of Flos Trollii had been established. Finally, the content of total flavonoids was compared in root,stem,leaf and flower of Trollius chinensis Bunge. Analytical results were as follows:The content of total flavonoids in the flower was up to 8.59%, 4.46% in the leaf, 1.42% in the stem, and the lowest in the root ,which was 1.27%.Considering comprehensive development of Trollius chinensis Bunge.,the leaf of Trollius chinensis Bunge.could be used as a medicine.The optimization of extraction technology in Floes Trollii. The optimization of extraction technology of the total flavonoids was carried out by orthogonal test and GA. With extraction yield and total flavonoids as the optimizing index,the optimum extraction procedures are as follows: 75% ethanol, 3-time extraction, 1.5h for each time by ethanol ,12 times volume solvent. The content of total flavonoids in the optimizing extraction process by ethanol was higher than that by water;but extraction yields by two methods were similar.The extraction yield was 44.31%, and the content of total flavonoids in Floes Trollii was 14.56%.Separation and combination of the the valid part in Flos Trollii. To select the optimum parameters for separating the total flavonoids in Flos Trollii, polyamide was used. Absorption and desorption capacities of three polyamides for total flavonoids were compared. The process condition such as the pH value of the sample solution, the concentration of the sample solution, the maximum absorption capacity, the eiuting reagent were ascertained. The optimum results were : Absorption and desorption abilities of 60~80 polyamide for total flavonoids were the best of three polyamides; The optimum pH value was 3, the concentration of the sample solution of Flos Trollii was 1g/3ml, the maximum absorption capacity for Flos Trollii was 0.2833g/ml, the eiuting reagent was water and 50% ethanol. The content of total flavonoids in the the valid part was 36.16% ,comparing 14.56 % of extraction substance.60~80 polyamide was studied as a good material of absorption and separation of total flavonoids in Flos Trollii under the technological conditions mentioned above.The study of antibacterial effect in vitro. The method of doubling delution in tube was used to detect the MIC and MBC of extracts to bacteria. Ethanol extraction technology had a good antibacterial effect.The Genetic Algorithm (GA) was used in the experiment. The study supplied a scientific research methodology for the effective substances of Chinese herbal medicines.
