Studies On The Purifying Techniques And Ertraction Quality Of Flavonoids In The Acrial Part Of Gentiana Scraba

Purpose:To establish a new analyzing method of total flavonoids and three marker components——an giferin,iso orient in and isovitexin in acrial part of Gentiana scabra,select the best process of extracting technology and purifying tech-nology and create a quality standards method of purified productsMethods:Using AlCl3 chromogenic method by visible spectrophotometry to prepare rutin standard curve in order to measure total flavonoids in acrial part of Gentina scabra.Using high performance liquid chromatography(HPLC)method with 0.1%formic acid-0.1%acetonitrile as elution system to establish standard curves of mangi feri n,isoorient i n and isovitexin so that we can de-termine the content of the three components respectively.Using extraction ratio of total flavonoids to optimize the extracting process.Using the adsorption rate and desorption rate to choose the best purifying process.Results:Established the content method of total flavonoids and other three marker com ponents——mangiferin,isoorientin and isovitexin by U V and HPLC re-spectively.The precision,repeatability and stability were good,the recovery was qualifiedSelected the best process o fextracting technology and purifting tech-nology.Firstly,16 times volume of 80%ethanol was put in the acrial part of Gent i ana scabra powder,ex traced 60 minutes for 2 times,then the products were pretreated before purified by AB-8 macroporous adsorption resin column.A fter 5BV water and 5BV 10%ethanol elution,7BV 50%ethanol elution liquid was collected and dried,then gained the pufified pfoduct.The total flavonoids content and trans fer rate were 52.63%and 80.3%In addition.the HPLC finger print of extraetion were obtained with 16 common peaks.The similarity of 10 batches acrial part of Radix Gentianae extracts were more than 0.99.The content of total flavonoids.mangiferin,isoorientin and isovitexin were 52.67%,9.85%,4.75%and 9.85%respectively.Conclusions:The established content method in this study is stable and relia-ble.Besides.the determined extracting and purifying process is controllable and stable,the quality control method is normal and feasible,the content of puri-fying process reached 50%.This study supplied a powerful evidence to the basic research of acrial part of Gentiana scabra.
