In this paper,an internal standard analytical method has been developed by ultrasonic extraction and reversed-phase high performance liquid chromatography(RP-HPLC). Caffeic acid was analyzed by the internal standard analytical method.Nitrobenzene internal standard was used for determination caffeic acid of taraxacum,The linear range of the method was 0.15-50μg/mL,R~2=0.9998;Coumarin internal standard was used for determination caffeic acid of another natural products,The linear range of the method was 0.5-100μg/mL,R~2=0.9999.The relative standard deviation(RSD)of determination was less than 3.3%(n=5)for all natural products.The recovery of caffeic acid was between 95.7-98.4%Accelerated solvent extraction(ASE)-microwave assisted extraction(MAE)-ethanol deposition-low temperature drying method was used to extracted pectin from tobacco.Use trifluoracetic acid(TFA)solution- p-aminobenzoic ethyl ester(ABEE)derivative-high performance liquid chromatography analysed galacturonic acid and monosaccharides in tobacco hydrolysate.Depending on the same analysis method compared the difference between the tobacco pectin and apple pectin hydrolysate.On this basis,calculated in the pectin content of tobacco.Galacturonic acid,glucose,mannose,xylose standard matters work curves respectively are:Y=6.0438X-0.0009,R~2=0.9998;Y=6.6714X+0.6811, R~2=0.9993;Y=7.5865X+0.0756,R~2=0.9997;Y=8.7248X+0.1004,R~2=0.9997.Tobacco pectin contented galacturonic acid 15.88%,tobacco contented galacturonic acid 20.22 mg/g,so tobacco contented pectin 12.73%.The relative standard deviation of determination was 1.34-2.98%.The recovery of galacturonic acid was 98.38%.In addition,this paper researched HPLC fingerprint of tobacco flavor.Especially focused on the stability of HPLC analysis.Four different samples retention time RSD were less than 1%,content RSD were less than 6%.This results meeting requests of less than 10%.The results in this paper obtained for the study of relationship between tobacco quality and tobacco chemical composition provided a basis for analysis.Caffeic acid and pectin extraction method for tobacco industry's handling of the waste of resources to provide useful reference.Basic research of tobacco flavor fingerprint provide a reliable analysis for prodct quality control.
