PartⅠExperimental study of rats with CRFObjective To establish model of CRF rats by Platt and observing their general condition, body weight, renal function, pathological changes of renal tissue.Methods 48 rats were randomly divided into normal group(12 rats), sham operation group(12 rats)and operation group(24 rats). We established the rat model of CRF by Platt. We observed their general condition and Scr,BUN, also observed the renal pathological changes by light microscope.Results①Body Weight: In body weight, there was statistical significance among three groups(F=11.303,P<0.05).There was not statistical significantce between normol group and sham operation group(P>0.05); there was statistical significantce between sham operation group and operation group(P<0.05); and there was statistical significantce among operation group itself(P<0.05). With the time lasting, body wight of operation group rats which was falling off was lower than sham operation group(P<0.05).②Scr,BUN: In Scr, there was statistical significance between normol group and sham operation group(F=6.283,P<0.05); and there was statistical significantce among operation group itself(P<0.05). With the time lasting, Scr of operation group rats which was stepping up was higher than sham operation group(P<0.05). In BUN, there was statistical significance between normol group and sham operation group(F=4.308,P<0.05); and there was statistical significantce among operation group itself(P<0.05). With the time lasting,BUNof operation group rats which was stepping up was higher than sham operation group(P<0.05).③There was renal tissue damage after 8 weeks in operation group. Conclusions We established successfully the rat model of CRF, which was the basis of our further research of correlation between iPTH and cTnI.PartⅡExperimental study on the correlation between iPTH and cTnI on CRF rats.Objective To approach the correlation between parathyroid hormone(PTH) and troponin I(cTnI) in chronic renal failure(CRF) rats, in order to provide theory for early diagnosis and therapy in CRF heart complications.Methods 16 model CRF rats were randomly divided into model group and treatment group, 8 rats for each group. At the same time, 8 rats which was left in sham operation group was as control group.Rocaltrol capsule was administrated (20ng/100g) by a stomach tube in treatment group, and same amount of teal oil was fed in model and control groups. After 45 days, we recorded general condition and body weight, dected Scr,BUN, and we also dected iPTH and cTnI by ELISA.Results①Body Weight: In body weight, there was statistical significance among three groups(F=18.515,P<0.05).There was statistical significantce between control group and model group(P<0.05); there was statistical significantce between model group and treatment group(P<0.05); and there was statistical significantce among model group itself and treatment group itself(P<0.05). With the time lasting, body wight of model group rats was falling off but treatment group was stepping up,which was higher than model group at the same time.②Scr,BUN: In Scr, there was statistical significance between model group and treatment group(F=4.846,P<0.05); and there was statistical significantce among model group itself and treatment group(P<0.05). With the time lasting, Scr of model group and treatment group rats were both stepping up, which model group's was higher than treatment group's(P<0.05). In BUN, there was statistical significance between model group and treatment group(F=9.516,P<0.05); and there was statistical significantce among model group itself and treatment group(P<0.05). With the time lasting, BUN of model group and treatment group rats were both stepping up, which model group's was higher than treatment group's(P<0.05).③iPTH: In iPTH, there was statistical significance between model group and treatment group(F=38.020,P<0.05); and there was statistical significantce among model group itself and treatment group(P<0.05). With the time lasting, iPTH of model group rats which was stepping up was higher than treatment group(P<0.05).④cTnI: In cTnI, there was statistical significance between model group and treatment group(F=32.085,P<0.05); and there was statistical significantce among model group itself and treatment group(P<0.05). With the time lasting, cTnI of model group rats which was stepping up was higher than treatment group(P<0.05).⑤Correlation between iPTH and cTnI: In scatterplot, there was linear correlation tendency between iPTH and cTnI in model group, analysis indicated r= 0.71, P<0.05,so we could consider there was linear correlation between iPTH and cTnI in model group. In scatterplot, there was linear correlation tendency between iPTH and cTnI in treatment group, analysis indicated r= 0.68, P<0.05,so we could consider there was linear correlation between iPTH and cTnI in treatment group.Conclusions①It is taken place secondary hyperparathyroidism in the early period of CRF, then cause pathological changes of heart complications.②cTnI is the early index of cardiac damage in CRF rats.③There is linear correlation between iPTH and cTnI ,so we should think highly of iPTH in the therapy of cardiac damage in CRF.
