Objective: Through the measurement of RCAS1 which was a receptor-binding cancer antigen expressed on SiSo cells secreted by bone marrow stromal cells of aplastic anemia (AA)mice, to understand the expression of RCAS1 in immune-mediated mice; to measure the apoptosis of normal bone marrow mononuclear cells(BMMNC) stimulated by supernatant of bone marrow stromal cells in AA mice,the RCAS1 secreted by macrophage cell lines and the apoptosis of normal BMMNC stimulated by supernatant of macrophage cell lines, evaluate the relationships between RCAS1 mRNA and cell apoptosis, explore the role of RCAS1 in the pathogenesis of AA.Methods:①Expression of RCAS1 mRNA in bone marrow stromal cells of AA mice and macrophage cell lines were measured by reverse transcriptase polymerase chain reaction(RT-PCR), the expression of RCAS1 mRNA in bone marrow stromal cells of normal mice was normal controls.②The apoptosis of BMMNC in normal mice stimulated by supernatant of bone marrow stromal cells of AA mice and macrophage cell lines were measured by flow cytometry technology , the apoptosis of BMMNC in normal mice stimulated by supernatant of bone marrow stromal cells of normal mice was normal controls.③Analyze the correlation between the expression of RCAS1mRNA and the cell apoptosis.Results:①The expression intensity of RCAS1 in bone marrow stromal cells of AA mice was(2.17±0.87),the normal controls was(0.96±0.22 ); the expression intensity of RCAS1 in macrophage cell lines was(1.98±0.24), the normal controls was (0.84±0.21);the groups of AA and macrophage cell lines were significantly higher than normal control(sP<0.01).②The apoptosis of normal BMMNC stimulated by supernatant of bone marrow stromal cells of AA mic(e8.85±1.05)% was significantly higher than normal control(s2.10±0.25)%(P<0.01); the apoptosis of normal BMMNC stimulated by supernatant of macrophage cell lines(16.66±1.97)% was also higher than normal controls(2.10±0.25)%(P<0.01).③Expression intensity of RCAS1 mRNA secreted by bone marrow stromal cells of AA mice was positively correlated with the apoptosis of normal BMMNC stimulated by supernatant of bone marrow stromal cells in AA mice(r=0.90,P<0.01),expression intensity of RCAS1 mRNA secreted by macrophage cell lines was positively correlated with the apoptosis of normal BMMNC stimulated by supernatant of macrophage cell lines(r=0.95,P<0.01).Conclusion: The expression of RCAS1 in AA mice and macrophage cell lines were high , RCAS1 maybe participate in hematopoietic failure of AA through the role of cell apoptosis in hematopoietic cells.
