Objective:To explore the relationship between immunotoxicology and kidney toxicity of the rat resulted in the clinical dosage of tripterygium Glycosides,to study the relationship between time and effect.Methods:Detect the basic immunological index and the general state of health inculding:thymus,spleen,immunoglobulin G(IgG)and immunoglobulin M(IgM), immune activity cells(natural killer cell,T-cell and B-cell activity )function.The urea,creatinine(Cr) and uric acid(UA);morphology of kidney were investigated too. Immunohistochemistry technique was employed to study the CD40 antigen at the macrophage and Fas expression in kidney,through ELISA to investigate the TNF-αin the homogenate of kidney.To investigated the urea,the creatinine(Cr),the uric acid(UA) and morphology of kidney for convalescence rats.Through analyzing the statistics result,to explore the relationship between immunotoxicology and kidney toxicity of the rat resulted in the clinical dosage of glycosides of tripterygium wilfordii hook f,to study the relationship between time and effect.Results:(1)The weight of experimental groups decreased significantly in the 5th group(p<0.05),the index number of the spleen decreased in the 5th group(p<0.05), the index number of the thymus decreased(p<0.05) in the 3rd and 4th group, significantly n the 5th group(p<0.01).The proliferation of experimental T and B cell decreased(p<0.05 )at 4th week;and decreased signifcantly(p<0.01) at the 5th week,the activity of NK decreased(p<0.05) at the 4th and 5th week in the experimental group. The content of IgG in the blood serum decreased(p<0.05)at the 3rd week,The content of IgG and IgM in the blood serum decreased signifcantly(p<0.01) at the 4th and 5th week.(2)The expression of the CD40 in the epithelium cell of renal tubule increased (p<0.05) at the 4th and 5th week.The expression of the TNF-αin the kidney increased(p<0.01) at the 4th and 5th week.The expression of Fas in the epithelium cell of renal tubule increased(p<0.05) at the 4th week,signifcantly(p<0.01) at the 5th week.(3)The urea,creatinine(Cr) and uric acid(UA) increased(p<0.05) at the 4th and 5th week.The kidney function of the 4th and 5th dministration group are almost normal(p>0.05),HE showed inflritation of netrophlic cell and lymphocyte in the renal tubule,the epithelium cell of renal tubule present degeneration,necrosis,Conclusion:(1)The tripterygium Glycosides can lead to immunotoxicology,the immune function and index of organ were depressant effect by Tripterygium Glycosides,and also concerned with the administration time.(2)The tripterygium Glycosides can r lead to kidney toxicity of the rat,and also concerned with the administration time too.(3)The following maybe is the reason for the mechanism of kidney toxicity.①The immunotoxicology of tripterygium Glycosides can increased the expression of the CD40 in the epithelium cell of renal tubule and TNF-αin the kidney,which can lead to kidney toxicity.②The immunotoxicology oftripterygium Glycosides can increased the expression of the Fas,make the epithelium cell of renal tubule apoptosis,which lead to kidney injury.③Tripterygium Glycosides maybe as antigen combines antibody to formed IC,injumed kidney.
