Traditional Chinese Medicine The Chuanxiong Serum Drug Chemistry And The Chemical Composition Of The Through The Blood-brain Barrier Research

The properties of Traditional Chinese Medicine(TCM)is the foundation and core of TCM. It generalized the character and function of Chinese medicine highly, excluding the fundamental substance and therapeutic function.So the research of active components should be guided by the theories of TCM, especially combined with the properties of TCM.Thus, in order to illuminate the fundamental substance about"upstream the head and downstream the lifeblood"guided by the theories of TCM, the analysis of constituents in blood and brain tissue after oral administration of Rhizoma chuanxiong have been carried out by using the method of Serum Pharmacochemistry of TCM and HPLC fingerprint. Then the compounds of effective fraction had been isolated and identified using the methods of column chromatograph and spectrophotometry. It groped for a new mode to research the foundation of active components, and would be promoted the development for assessment of Rhizoma chuanxiong quality. The contents are as follows:1 Serum Pharmacochemistry of the ethanol extract from Rhizoma chuanxiongThe serum fingerprint spectrum of the ethanol extract from Rhizoma chuanxiongwas established after confirming methanol method to prepare serum samples and the time of gathering blood samples after oral administration of the ethanol extract ninety minutes as optimal time. There were fourteen peaks which found in all the serum fingerprint spectrum. Meanwhile, adopted the spectrum analysis and the literature information, chemical structural of the thirteen constituents absorbed into blood were identified. The compounds butyraldehyde,3-butylphthalide,3-butylidenephthalide,senkyunolide A,neocnidilide,Z -ligustilide,Ferulic acid originally existed in the Rhizoma chuanxiong.2 Analysis of the chemical components which penetrated the blood brain barrierAdopted the spectrum analysis of HPLC and GC/MS, seven chemical components were identified through the comparison between the brain tissue before oral administration and after oral administration. The compound included diethyl phthalate,Palmitic acid,Stearic acid originally existed in the Rhizoma chuanxiong.3 Isolation of the active compouds from Ligusticum chuanxiong Hort.The ethyl acetate extracted fraction could restrain the conglomeration of blood platelet according as the dose,which could be indicated it was the effective fraction. By the means of systematic solvent extraction, column chromatograph and re-crystallized, 3 compounds have been gained by separated and purified from the ethyl acetate extracted fraction of Rhizoma chuanxiong. Adopted the spectrum analysis and the literature information, chemical structures of 3 components have been identified, which are Ferulic acid, Caffeic acid, Vanillic acid.4 Reasonability of total ferulic acid for quality assessment of Rhizoma chuanxiong and determination of total ferulic acid by HPLCFounded on the literature, it was found that the determination of ferulic acid could rise after storing for several years. The main reason was that coniferyl ferulate in Rhizoma chuanxiong was found liable to hydrolyze into ferulic acid. Therefore, we discussed the reasonability of total ferulic acid for quality assessment of Rhizoma chuanxiong using a gradient program by HPLC-DAD and developed created a new HPLC method to quantitatively analyze total ferulic acid in Rhizoma chuanxiong. It would be referenced for improving the quality standard of Rhizoma chuanxiong.
