For the globalization and modernization of Chinese herbal medicine,it is indispensable to improve the quality control process.Ambient ionization mass spectrometry is a high-throughput and low-cost technology,and has gradually shown its advantages in the quality control of traditional Chinese medicine.Ligusticum chuanxiong as a frequently-used Chinese medicine has a mixed authenticity in the market.This paper takes Ligusticum Chuanxiong as the research object and realizes the rapid identification of samples by PS-MS technology.The specific research contents are as follows:1.Introduction.This section summarizes the research progress of Ligusticum chuanxiong,including the an overview of Ligusticum chuanxiong,its identification and quality control methods.The extraction methods of modern Chinese medicine are further elaborated,followed by the influence of processing of Ligusticum Chuanxiong on the final medicine's chemical composition and its function.This section ends with a summary of the development status of open-ended mass spectrometry.2.Study on the identification of Ligusticum Chuanxiong by HPLC fingerprint method: the high-performance liquid chromatography fingerprints of Ligusticum Chuanxiong is established using high performance liquid chromatography-mass spectrometry(HPLC-MS)based on standard product.The HPLC fingerprints of 25 batches of genuine Ligusticum Chuanxiong and 10 batches of Rhizome ligustici are analyzed by ChemPatternTM metrological software.The similarities of Ligusticum Chuanxiong and Rhizome ligustici with common patterns are between 0.85-0.99 and 0.35-0.45,respectively.The scores of PC1 and PC2 of Ligusticum Chuanxiong and Rhizome ligustici are 76.81% and9.36%,respectively.These measured values clearly divide into two categories,which proves that the authenticity of Ligusticum Chuanxiong can be successfully identified by the fingerprint of HPLC.This research provides a basis for the open-ended mass spectrometry analysis of Ligusticum chuanxiong.3.Identification of Ligusticum Chuanxiong by PS-MS: PS-MS fingerprint profiles of Ligusticum Chuanxiong and Rhizome ligustici are studied.The main differences are identified through comparison.The taper hole voltage and spray voltage of PS-MS under positive and negative ion mode are optimized.The stability and repeatability of the method are examined by ChemPatternTM metrology software.The PS-MS fingerprints of 25 batches Ligusticum Chuanxiong and 10 batches of Rhizome ligustici are analyzed.The similarities of Ligusticum Chuanxiong and Rhizome ligustici with common pattern are 0.60-0.90 and 0.75-1,0.30-0.40 and 0-0.25,respectively.The PCA scores of Ligusticum Chuanxiong and Rhizome ligustici are 4.14% and 9.44% and44.23% and 63.52%,respectively.The measured values divide into two categories.The results of identification are consistent with those of fingerprint analysis by HPLC.This method is simpler,faster and with a lower cost as compared to the fingerprint method of HPLC.4.Study on quality control of Ligusticum Chuanxiong processing based on HPLC and PS-MS: Because liquor-making Ligusticum Chuanxiong is widely used in clinic,the liquor-making technology of Ligusticum Chuanxiong is chosen as the targeting object of this study.The contents of four main active substances in Ligusticum Chuanxiong before and after processing are quantitatively compared using HPLC technique.The results suggest that the content of ferulic acid and ligustilide,the main active ingredients of Chuanxiong Liquor,increased significantly,while the content of pine cypress ferulate and ligustilide A decreased drastically.The optimum processing conditions discovered in this study are as follows: the content of yellow rice wine is 30%,the baking temperature is 100?,and the baking time is 2 hours.The fingerprint of HLPC and the chemical profile of PS-MS of Ligusticum chuanxiong's raw and wine products are established.The fingerprints of raw and processed products of Ligusticum Chuanxiong are established by PS-MS technology.The results reveal that the fingerprint of Ligusticum Chuanxiong produced by wine and raw products could not be identified by HPLC.However,PS-MS chemical contour methodcould be adapted for the identification of Ligusticum chuanxiong,which shows its superiority in rapid analysis of Ligusticum chuanxiong.
