Three species of Compositae systematically for scientific development and utilization of naturally active components from medicinal plants growing in ethnic area in China were studied.This thesis primarily described the phytochemical investigation of the whole plant of Senecio scandens var.incisus and Ligularia duciformis which were herborized in MeiGu county of Sichuan Province along with Ligularia hodgsonii collected in Liangshan,the northeast part of Liangshan Yi Autonomous Prefecture of Sichuan Province.Twenty compounds were isolated by means of silica gel column chromatography, Sephadex LH-20 column chromatography and preparative thin layer chromatography.Thirteen compounds were elucidated on the basis of spectroscopic methords(UV,IR,EI-MS,HR-ESI-MS,1HNMR,13CNMR and 2D NMR(1H-1H COSY,HMQC,HMBC,NOESY),and by comparison with the data from literatures,five of which were new compounds.A new jacaranone derivative 2,3-dihydrogen-3-hydroxy-jacaranonoic acid(1) has been isolated from the whole plants of Senecio scandens var.incisus,together with three known compounds:2"-(p-hydroxyl-cinnamoyl) -6"-jacaranone-D-glucopy-rano-side(2),2"-caffeoyl-6"-jacaranone-D -glucopyranoside(3) and kampferol-3-rhamnoside(4).Compounds 2-4 were isolated from the plant for the first time.Meanwhile,four new sesquiterpenoids,3β-acetoxy-6β-methoxyeremophil-7(11),9(10)-dien-8β,1 2-olide(1),3β-acetoxy-8α-hydroxy-6β-methoxyeremophil-7(11),9(10)-dien -8β,12-olide(2),3β-acetoxy-10β-hydroxy-6β,8β-dimethoxyeremophil -7(11)-en-8α,12-olide(3) and 3β-acetoxy-6β,8β,10β-trihydroxyeremophil-7(11) -en-8α,12-olide(4) with a known 3β-acetoxyl-8β,10β-dihydroxy-6β-methoxyeremophilenolid (5) were isolated from the methanol extract of whole plants of Ligularia duciformis.Additionally,four known compounds Quercetin(l),(E)-3-(2-hydroxy-4-methoxyphenyl)-2-acrylic acid(2),(E)-3-(2,4-diriydr-oxyphenyl)-2-acrylic acid(3) and(S)-3-hydroxy -3-phenylpropanoic acid(4) were isolated from Ligularia hodgsonii,.These four compounds were also isolated from the plant for the first time.The cytotoxicity of the new compounds against human pneumonic cancer cells A-549,human hepatic cancer cells Bel-7402 as well as human colonic cancer cells HCT-8 were evaluated by using MTT method, the results showed they have no cytotoxicity against the three tumor cells.Results of this thesis provide scientific data to the conservation and sustainable use on plant resources in ethnic areas.Chemical composition and pharmacological research of Ligularia from 2003 to 2008 is also reviewed.
