Purpose:To investigate the reconstruction capacity of Slvia milt-iorrhiza and rabbit bone marrow-derived mesenchymal stem cells(MSCs) venous injection on the infarcted myocardium after acute myocardial infarction,and further research the effects on the infarcted myocardium Gap junction expressions.Material and method:Firstly,Bone marrowwas extracted from rabbit's tibia on asepsis condition,and then MSCs from rabbit were isolated,cultured,amplificated and marked with BrdU.The model of acute myocardiac infarction was established by ligatvlg animal left anterodecendant arteries(LAD).The rabbits that Myocardial infarction model has been established were randomly divided into 3 groups,the model group,venous injection Slvia miltiorrhiza and MSCs group,venous injection MSCs group.One week after,the group that venous injection MSCs were given BrdU-labeled MSCs(2×10~6) through auris vein,and the group that venous injection Slvia miltiorrhiza and MSCs were given BrdU-labeled MSCs(2×10~6),simultaneity were given Slvia miltiorrhiza (15.4mg/kg) through auris vein for 10 days.Five weeks after,Infarct area Brdu immunohistochemical staining to observe whether MSCs Point transplantation moved to the infarct zone;Brdu staining positive-stained cells of cardiac-specific troponin T staining to observe the migration and differentiation of cells and the nature of differentiation;The expression and distribution of Cx43 was observed by the RT-PCR method;the changes of myocardial ultrastructure was observed by transmission electro microscope.Results:1.There were Brdu positive cell in infarct area of two group that the venous injection Slvia miltiorrhiza and MSCs group and venous injection MSCs group,and did not detect in infarct area of the model group.In addition,anti-BrdU stain positive cells expressed Tropnin-T.2.Cx43 were expressed in infarct area of two group that the venous injection Slvia miltiorrhiza and MSCs group and venous injection MSCs group by RT-PCR, and did not detect in the control group.In addition, venous injection Slvia miltiorrhiza and MSCs group had significant differences in cardiac function compared with venous injection MSCs group(P<0.05).3.The myocardial ultrastructure of venous injection Slvia miltiorrhiza and MSCs group were observed that it is similar to normal myocardial ultrastructure by electromicroscope,but the myocardial ultrastructure of venous injection MSCs group were observed that there are obviouss changes by electromicroscope.Conclusion:1.Through Slvia miltiorrhiza and rabbit bone marrow-derived mesenchymal stem cells(MSCs) venous injection on the infarcted myocardium after acute myocardial infarction,MSCs can be moved to the infarct zone,and can differentiate into myocardial cells,throngh venous injection Slvia miltiorrhiza and MSCs.2.It is obvious that Slvia miltiorrhiza could ameliorate the Gap junction of new myocardial cells in the infarcted myocardium after acute myocardial infarction,decrease the expression of Cx43 evidently,and the ultrastructure of heart and expression of Cx43 in treated group were approach to normal group. Accordingly,it lays foundations for new myocardial cells have normal function.
