| Objective:Chronic pancreatitis is due to the biliary disease or alcohol poisoning in the pancreas impairment and fibrosis, often accompany calcification, pseudocyst, the diminution or atrophy of islet cells.Its pathophysiology is complex, the characteristic is too much sedimentation by fibroblasts and Extracellular matrix(ECM)in the pancreas.The traditional index of pancreas fibrosis are the rising of the Laminin(LN)level and the decrease of the superoxide dismutases (SOD) activity.The Laminin (LN) level and the superoxide dismutases(SOD)activity were detected in normal group(pancer fibrosis group),control group(Chronic pancreatitis group),interference group(small dose Astragalus injection group),interference group(medium dose Astragalus injection group)and interference group(large dose Astragalus injection group)by using enzyme immunoassay (ELISA),and the pathological change of pancreatic tissue was observed. To investigate the therapeutic effect of Astragalus injection on Chronic Pancreatitis.Methods:To choose SD rats,fractionate into 5 group randomly,there are 10 rats in each group.They are normal group,control group,mall dose Astragalus injection interference group(400mg/kg),medium dose Astragalus injection interference group(800mg/kg)and large dose Astragalus injection interference group (1600mg/kg).Except normal group,the other 4 group rats were preoperative fasting 12h,Amiodarone(100 mg/kg weight)intraperitoneal injection of anesthesia,made midline incision along linea alba,occlusion pancreatic bile ducts near porta hepatic,through constant flow pump injection TNBS 2% ethanol phosphate buffer 0.4ml to carunculae major,in 1h, to prevent drugs,after closing constant flow pump pressure maintaining 10min,procedure was strict aseptic operation.In order to prevent the rat dehydration,we subcutaneous injections physiological saline 6ml to rats, two times,until the rats can freely drinking.Regular diet after postoperative fasting 72h.Using different doses of Astragalus injection intramscular injection the rats after 28 days.All observed object were taken suction venous blood, to separate blood serum,put them to-20℃deep freeze refrigerator to reserve.We put the blood serum to room temperature before the experiment,made them centrifugal,took supernatant to determination.The Laminin(LN)level in normal group,control group and interference group was detected by using enzyme immunoassay(ELISA).SOD total activity was detected by xanthine oxidase.The change of chronic pancreatitis tissues fibering was observed by using HE staining.Results:The pathological change of pancreatic tissue was observed in control group rats. Model rats 4 weeks after modeling, showed interstitial inflammatory cell infiltration of pancreatic, canal of Wirsung fibroplasias,canal of Wirsung hyperplasy and broaden,fibrosis and inflammatory cell infiltration in canal of Wirsung,pancreatitis lobule,part of glandular organ was atrophy and necrosis,gland alveolus turn to vacuole.Fibrosis degree of Astragalus injection interference group significantly reduced than model group.Giving pathological section of five group rats's pancreatic tissue fibrosis scores,find the level of pancreatitis fibrosis in model rats was much higher than it in interference control. SD rats normally compared with the control group's,control group rats the pancreas fibrosis significantly,LN content increased significantly,SOD activity reduced significantly.The level of pancreas fibrosis reduced in Astragalus injection interference group,the LN level reduced significantly and the SOD activity increased significantly in Astragalus injection interference group.Conclusions:Astragalus injection has the role in the improvement of rat pancreatic fibrosis. Through the treatment of Astragalus injection to rat chronic pancreatitis fibrosis, we observed that the level of Chronic pancreatitis fibrosis pancreatic tissue fibrosis reduced obviously from pathological changes,the LN content reduced and the SOD activity increased significantly in Chronic pancreatitis blood serum. |
