Extraction craft conditions were investigated by applying orthogonal test and method of quality control of Compound dangui granules was established by HPLC.Firstly, according to the request of process planning, extraction craft conditions of naphtha were investigated and the identified optimal craft was A1B2C2 (soaking for an hour, eight times volume of water, extracting for five hours) by regarding the content of naphtha as index. Secondly, the including methods of naphtha were investigated and the identified optimal craft was A1B3C2D2 (the ratios of the volume of water toβ-cyclodextrin and the amount of naphtha toβ-cyclodextrin were 10:1 and 1:8 respectively, stiring time and temperature were 2 hours and 40℃respectively) by regarding the inclusion rate and the yield of inclusion compound as index. Thirdly, ethanol extraction craft and water extaction craft were optimized by regarding the contents of Astragalus Saponin I and Salvianolicacid B as the indexes, the optimal ethanol extraction craft and water extaction craft were A1B1C2D3(eight times volume of 50% ethanol,extracted for 3 times by 1.5 hours per time) and A3B1C3(twelve times volume of water,extracted for 3 times by 0.5 hours per time) respectively. Finally, the orthogonal test was used and determined the optimal clearification process was A1B2C1A3(5% ZTC-Ⅱclarifier was added to the concentrate, the ratio of medicine to liquid was 1:2, the experimental temperature was 50℃) by regarding extraction ratio as index. The above studies provided reference frame for the determination of the extraction craft of Compound dangui granules.The therapeutic action of Compound dangui ganules were investigated by pharmacodynamics. Results indicated that Compound dangui ganules could improve biochemical indicator in blood and urine of CRF rat and hepatic tissure conformation, the level of usea nitrogen and creatinine in the serum and Urine protein decreased significantly. Results of acute toxicity testing indicated that the maximal tolerance dose (MTD) of Compound dangui ganules was 60g/kg and no obvious sign of acute toxicity was observed.Antioxidant activity of Compound dangui ganules was investigated by the method of DPPH. Compound dangui ganules was compared with niaoduqing ganules and negative control, As a result, the antioxidant activity of Compound dangui ganules was stonger than which of niaoduqing ganules. At the same time, the antioxidant activity of the water-soluble costituents of Radix Salivae Miltiorrhizae were investigated by applying vitamin C for reference and results showed that the antioxidant activity of Salvianolicacid B was the strongest, furthermore stronger than vitamin C, the antioxidant activities of Protocatchuic acid and Ferullic acid were close to each other.Furthermore, The comparison of different extraction methods of Radix Salivae Miltiorrhizae was carried out by the determination of the content of Salvianolicacid B by hign-performance liquid chromatography. As a result, after ultrasonic extrction for 10 minutes by 75% methol, the content of Salvianolicacid B equated to that extracted by the method of refluxing extraction in Chinese pharmacopoeia 2005. The extracted method is very simple and save time. Finally, HPLC-ESI-MSn was adopted to analyse the water-soluble constituents of Radix Salivae Miltiorrhizae and seven compounds were identified.
