The Effects Of Compound Danshen Decoction On Cyclophosphophamide Model Of Mouse Bone Marrow Inhibition

Objective:Observating the effect on model in mice using cyclophosphophamide peripheral target.At the same time,detecting of bone marrow hematopoietic stem cell content and the change of bone marrow cell cycle.To study the effect of Danshen Decoction on mice hematopoietic stem cell.To explore the tumor patient's teatment after chemotherapy and provide the basis of Chinese medicine,also explore the effect of Danshen Decoction on hematopoietic stem cells.Methods:Mice were randomly grouped and divided into physiological control group,Danshen group and normal group.There were 25 mice in each group.Experimental treatment group were given a variety of traditional Chinese medicine decoction enrichment 0.25ml/d orally,once daily,for 28 days.Control group were given equivalent distilled water orally,once daily,for 28 days.Excluding normal group,at the first 9,11 and 13days,the remaining groups of mice were given cyclophosphophamide 100mg/kg by intraperitoneal injection.10 mice were randomly selected in the cyclophosphophamide group,peripheral blood was checked by using eyeball method,disposed at the anticoagulation tubes.The remaining mice in the experiment were put in the same way and peripheral blood was put in anticoagulation tubes,too.Two groups were used to detect the peripheral blood WBC,RBC,HGB and PLT,and to compare the analysis of their differences.10 mice were randomly selected in the cyclophosphophamide group,killed by cervical dislocation method,bilateral femoral were disposed, repeatedly washing with normal saline by 6^th needle out of all the femoral bone marrow.The femoral bone marrow was centrifuged for 10 minutes at 2000r/min,leaving supernatant,adding 4ml cold 75%ethanol fixed,shaked by shaking machine.The same bone marrow samples were produced from the remaining mice in the experiment at the 28^th days.The bone marrow hematopoietic stem cells and the cell cycle of two groups were detected and to compare their differences.Results:1.There was no difference between physiological control group and Danshen Decoction group in WBC,RBC and PLT,bone marrow CD34⁺ hematopoietic stem cells.Hoewer,peripheral WBC,RBC and PLT,bone marrow CD34⁺ hematopoietic stem cells was lower than those of the normal group.2.After the experiment,the peripheral WBC,RBC and PLT,bone marrow CD34⁺ hematopoietic stem cells of Danshen Decoction group was increased more than those of normal physiological control group.There is significant difference(P＜0.05).3.The increased percentage of peripheral blood WBC,RBC,PLT and bone marrow CD34⁺ hematopoietic stem cells was more than which of physiological control group.Conclusions:1.Compound Danshen drink can stimulate peripheral blood WBC,RBC and PLT of bone marrow suppression mice.2.Compound Danshen drink can stimulate CD34⁺ hematopoietic stem cells of bone marrow suppression mice.3.Compound Danshen drink can become a possible tumor patient's treatment after chemotherapy with Chinese medicine proram.
