Objective:To study the effect of coprinus comatus polysaccharide on humoral immune function of immunosuppressive mice,to further its development of an immune modulator to provide experimental evidence.Methods:Kunming mice, male and female in half, Were randomly divided into normal group (A),model group (B),mushroom polysaccharide group (C),low-dose CCP group (D),medium-dose CCP group (E),and high-dose CCP group (F), in total six groups. Except from group A,the remaining mice in each group were prepared with Cy into immunocompromised animal model. Eight days later, give medicine to the mice of each group in the early morning. Give group C with 5mg/kg lentinan, group D 225mg/kg CCP, group E 450mg/kg CCP, group F 900mg/kg CCP,at the same time, give group A and B normal saline,Give medicine to them once a day for 14 days. Weight mice in each group the day after withdrawal,take blood sampling after eyeball were extirpated, and then kill them. Extirpate thymus and spleen and quantify wet weights, at the same time calculate the thymus index and spleen index;In this experiment,detect proliferation of B lymphocyte throught MTT ;mensurate content of serum hemolysin to detect the level of antibody- secreting;using quantitative hemolysis method to detect effects to forming antibody cell;levels of IL-2,IL-4 in serum of mice in each group was detected by ELISA method.Results:1.Thymus index and spleen index of mice in each group:compared with normal group, the spleen index of model group was significantly reduced(P﹤0.05);compared with the model group, spleen index of immune suppression mice in other different doses CCP groups were significantly increased, the difference is statistically significant(P﹤0.05),while no significant effect on thymus index.2.The B lymphocyte proliferation rate of mice in every group:compared with the normal group,model group B-lymphocyte proliferation rate significantly reduced(P﹤0.05);compared with the model group, B lymphocyte proliferation rate of the mice in medium-dose and high-dose CCP group has increased significantly, the difference is statistically significant(P﹤0.05).3.The amount of secreted antibody of mice in each group:compared with normal group, the amount of secreted antibody of model group mice significantly reduced(P﹤0.05);compared with the model group, the secreted antibodies amount of the mice in different dose CCP groups has increased significantly, the difference is statistically significant(P﹤0.05).4.The number of forming antibody cells of mice in each group:compared with normal group, this number of mice in model group significantly reduced(P﹤0.05);while this number of different dose CCP groups increased significantly, the difference is statistically significant(P﹤0.05).5.IL-2, IL-4 levels in Serum of mice in each group:these two levels of mice in model group were significantly lower than the normal group(P﹤0.05); compared with the model group, the medium-dose and high-dose CCP group mice serum IL-2 and IL-4 levels increased significantly,the difference is statistically significant(P﹤0.05).Conclusion:1.Different dose CCP can increased spleen index of immunosuppressed mice, but no significant effect on thymus index.2.Medium-dose and high-dose CCP can increase B-lymphocyte proliferation rate of immunosuppressed mice.3.All different dose CCP can increase the amount of antibody- secreted in immunosuppressed mice.4.Different dose CCP can increase the number of new forming antibody cells in immunosuppressed mice.5.Medium-dose and high-dose CCP can increase the level of IL-2 and IL-4 in serum of immunosuppressed.
