Coix Seed Processing Technology And Quality Standards To Standardization Study

Purpose:Coix seed is the dried and mature kernel of jobstears,which belongs to Gramineae. Coix seed has the effectiveness on invigorating the spleen, promoting urination and removing heat to eliminate pura. Coix seed processed by stir-baking with bran is main processed product in ,but its procedure is indefinite.Its quality standard and processing affect has not reports also.So the topic is to optimize the procedure of Coix seed, compare the effect on excreting dampness and invigorating the spleen of before and after procedure,establish the quality criteria of Coix seed and its processed product. Material and method:Materia:Coix seed purchased from Anhui Jiren Pharmaceutical Co., Ltd. ,identify by ZHAI Yan-jun, Professor who belongs to staff room of the identification of chinese materia medica which belongs to Liaoning University of Traditional Chinese Medicine, is the dried and mature kernel of jobstears,which belongs to Gramineae.Method:1.Serving the glycerol trioleate and polycose as the index,acting the quantity of bran,the temperature of procedure and the time of procedure as factor of investigation,the procedure was optimazed by orthogonal design.2.Thin-layer chromatography, gas chromatography and liquid chromatography were compared before and after the changes of the coix seed.3.The rats of hyperlipemia were lavaged for two weeks by the suspl of coix seed and its processed product and the effective on lowering blood-fat was compared by determining the content of TC,TG,LDL and HDL.4.The mouse of splenic asthenia were lavaged by extract of each fraction of coix seed and its processed product and the effective on invigorating the spleen was compared by determining gastric emptying and facilitation of bowel.5. Serving the glycerol trioleate and polycose as the index,the quality criteria of coix seed and its processed product was established.The impurity,moisture content,ash content and extractive of the coix seed and its processed product of ten habitats were determined.Results:1.The optimal procedure was 100g coix seed that the temperature was 210 to 220,time was 60 seconds and the quantity of bran was 100:20,optimized by orthogonal design.2.The coix seed was different from its processed product on both quality and quantity ,and a new chemical constituent was separated and identified as 5-HMF,observed by varied chromatography of the coix seed and its processed product.3.The coix seed and its processed product had an effective on lowering the content of TC,TG,LDL of rats,steping up the content of HDL of mouse,lowering the content of blood-fat,including the effective of coix seed surpassed its processed product.4. The coix seed and its processed product had an effective on depressing down gastric emptying and facilitation of bowel,including the effective of its processed product surpassed coix seed.5.The technology of glycerol trioleat and polycose consistented with relative regulation.The content of polycose of coix seed was tentatively set on more than 2.0%,the content of glycerol trioleat was more than 0.5%;the content of polycose of its processed product was more than 3.5%,the content of glycerol trioleat was 0.7%.Conclusion:1.The optimal procedure of processing drugs is determined and the scaling-up is carried out,the result is the basic same as subsample's.The method of stir-baking with bran of coix seed is stable and feasible.2.The change happens on quality and quantity of before and after procedure of coix seed and 5-HMF,a new chemical compound,is identified after procedure.Its lead to the changes of the functions.3. The coix seed and its processed product had an effective on lowering the content of blood-fat,including the effective of coix seed surpassed its processed product.It suggest that coix seed used descending blood-fat better than proceed in clinical.4. The coix seed and its processed product had an effective on invigorating the spleen, including the effective of its processed product surpassed coix seed. It proved that coix seed proceed with stir-baking used in invigorating the spleen in clinical is significanced. 5.The impurity,moisture content,ash content and extractive of the coix seed and its processed product of ten habitats were determined and the quality criteria of coix seed and its processed product is stable and feasible.It can be used to evaluate and control the quality of the coix seed and its processed product also.
