Photosynthetic bacteria (PSB) produces hydrogen under anaerobic and light-illuminated conditions. High combination of organic pollution removal and hydrogen production made PSB to be a new hot spot in bio-hydrogen production field. This paper regarded PSB group selected as the main research object and the properties of hydrogen production were explorated. The innovation of this paper was to present a new research of PSB hydrogen production through photosynthetic processes using hydrolyzate of corn stalks. This study was undertaken to open up a new way that can be followed for photosynthetic bacteria hydrogen production.PSB group were separated from garden soil. PSB group were cultivated with intermittent mode. The influencing factors including carbon sources, nitrogen sources, combination of carbon and nitrogen sources, initial pH value, illumination pattern, inoculation amount, light intensity, incubation temperature were discussed respectively. The results showed that carbon sources had markedly influences on hydrogen production, compared with nitrogen sources, the optimal carbon and nitrogen source were glucose and ammonium oxalate respectively.The best carbon and nitrogen source combination were glucose and sodium glutamate. The hydrogen production in acidity environment was higher than that in alkalinity. The hydrogen production quantity using the illumination mode of 12 h light-12 h dark alternation was higher than that using the illumination mode of continuous light.The group became dominant species and got the highest hydrogen production under the condition of inoculation amount of 10%, light intensity of 7000 lux, incubation temperature of 30℃.The influence factors of hydrogen production were investigated by experiment of orthogonal experiment. The optimum process conditions were as follows: the best bacteria group was number 10; the glucose was carbon source and concentration of carbon source was 3 g/L; the urea was nitrogen source and concentration of nitrogen source was 9 g/L; the inoculation amount was 10%; initial pH value was 8.5; the illumination mode was 12 h light and 12 h dark alternation; incubation temperature was 30℃. The main factors affecting hydrogen production were strain, carbon sources, carbon source concentration and nitrogen sources.At frist corn stalks were hydrolyzed, then hydrolyzate was detoxified. The effects of corn stalks hydrolysis conditions, hydrolyzate detoxification methods, nitrogen sources, phosphorous element, NaCl, NaHCO3 and metal ions on hydrogen production were investigated. The optimum hydrolysis process conditions were as follows: the sulfuric acid concentration was 1%, the solid-liquid ratio was 1:12, the hydrolysis time was 0.5 h and the hydrolysis temperature was 110℃, respectively. There hydrolyzate detoxification methods were compared and the optimal method were calcium. The maximum highest hydrogen production was obtained respectively under the condition of urea 2 g/L, phosphorous element 0.05 g/L, NaCl, 1.5 g/L, NaHCO3 10 g/L. The the most significant factors was NH4+. The results showed that photosynthetic bacteria group using crops as substrates for hydrogen production is a low cost and high efficiency hydrogen production technology. It will play an important role in bio-hydrogen production field.
