| Auricularia Auricula is an important kind of edible fungi, Polysaccharides, one of main active components of A. auricula, have various beneficial to human body functions such as reducing blood lipid antitumor, anticoagulant, etc..The paper has extraction got water-soluble and alcohol-soluble Auricularia auricular Polysaccharide (AAP) from A. auricular, using its fruit as rough material , and extraction agent, using water and 75 percents of alcohol.In this experience, four kinds of grafted monomer,acrylic ( AM ) ,acrylic acid(AB),acrylontrile(AN) and Poly Vinyl Acetate (PVAc) were grafted with water-soluble and alcohol-soluble AAP respectively.The results showed that the AM and PVAc was respectively grafted to water-soluble and alcohol-soluble AAP ,using ammonium persulphate-Sodium Bicarbonate as the initiator, by which the Auricularia auricular Polysaccharide gel membrane was prepared. And the influence of the reaction conditions on the water-soluble AAP grafting AM and the alcohol-soluble AAP grafting PVAc were studied and analyzed through the reaction time, the reation temperature, the reation pH, the amount of initiator, and the monomer concentration tatio, and so on. On the basis of single factor experiment established by the response surface optimization experiment model conclusion: the grafted ratio of water-soluble AAP grafting AM was the highest for 72.4395%, under the condition of temperature 50.37 , time 2h, proportion of alcohol-soluble AAP 21.5%, initiator of AM 3.68%, pH 6.04. The verifying test results indicated that the model was satisfactory.The grafted ratio of alcohol-soluble AAP grafting PVAc was the highest for 39.1794%, under the condition of temperature 57.03 , time 2.65h, proportion of alcohol-soluble AAP 26.36%, and initiator of PVAc 4.5%. The verifying test results indicated that the model was satisfactory.The two graftomers were respectively used to manufacture the 1 mm thickness of the membranes, meanwhile, the infrared structure of both kinds of grafted polymer was characterized. The results showed that graftomers had been introduced into the main chain of polysaccharide. The physicochemical properties of graftomer of gel membrane were studied through water imbibition, viscosity, membrane morphology.The results showed the physicochemical properties of water-soluble AAP grafting AM—water imbibition, viscosity, membrane morphology were better than that of alcohol-soluble AAP grafting PVAc.In addition, X-rays radiation shiedling effect of Polysaccharide gel membrane was also studied, based on the cell count experiment, DNA-Ladder experiment and MTT cell experiment. The results showed that all these two graftomers could reduce the damage degree of cell and DNA caused by X-rays radiation,and the protective effect on DNA protected by alcohol-soluble AAP grafting PVAc was better than that of water-soluble AAP grafting with AM. The results showed that both of the two polymers could fight against the X-ray. |
