Objective: To observe the effects of Galectin-3 (Gal-3) on proliferation and angiog- enesis of endothelial cell differentiated from bone marrow mesenchymal stem cells. It may be worthwhile to examine whether the angiogenic- promoting activity of the Gal-3 can be used as a therapeutic adjuvant strategy in some ischemia-related diseases and tissue engineered vascular grafts,and may provide a experimental basis for Bone marrow MSCs in vascular surgery basic research and possible clinical application.Methods: The cultured Mesenchymal stem cells were isolated and purified from bone Marrow of Sprague-Dawley rats by gradient centrifugation with lymphocytes separation medium. Cells of passage 3 were differentiated into endothelial cells by 10 ng/ml vascular endothelial growth factor(VEGF) and 2ng/ml basic fibroblast growth factor(bFGF). These cells were identified as endothelial cells by immunohistochemistry staining and electronic microscopy to observeⅧfactor associated antigen expression and ultrastructural characteristics after 14 days. The cells were cultivated with the Gal-3 at the concentrations of 0.1μg/ml,1μg/ml and 5μg/ml for 24 hours.1. The proliferation of endothelial cells differentiated from bone marrow Mesenchymal stem cells were measured by MTT ;2. Flow cytometry was applied to observe cell cycle; 3. The functionality of angiogenesis was observed when the cells showed the ability of tube formation in presence of Gal-3.Result: 1. The absorbances in the Gal-3 groups (1μg/ml and 5μg/ml) were 0.3002±0.0159 and 0.3514±0.0133, respectively, which were significantly greater than (P<0.05) that in the control group(0.2339±0.0041);2. Flow cytometry detection showed that S phase cells (%) are 29.42±0.45, 34.56±0.82 and 52.58±2.84 in groups of 0.1μg/ml, 1μg/ml and 5μg/ml, which higher remarkably compared with the control group (P<0.05); G2M phase cells increased in the concentrations of 1μg/ml(4.88±1.12) and 5μg/m(l5.26±0.45)(P<0.05) compared with control group(P<0.05);Proliferation index(PI)are 32.45±0.94, 39.44±0.75 and 57.84±2.52 in groups of 0.1μg/ml, 1μg/ml and 5μg/ml, and they increased remarkably compared with control group (P<0.05)3. The relative total length of the tubular network formation increased Significantly compared with control group (P<0.05).Conclusion: 1. Gal-3 can promote the proliferation of endothelial cells differentiated from bone marrow Mesenchymal stem cells.2. Gal-3 can promote the angiogenesis of endothelial cells differentiated from bone marrow Mesenchymal stem cells .
