Objective:To investigate the alterations in the maternal concentration of soluble endoglin(sEng)and transforming growth factor-β1 (TGF-β1)in pregnancy-induced hypertension disease (HDCP) patients.To explore the correlation of TGF-β1 and sEng with the pathogenesis of HDCP.Material and Methods:200 cases pregnant women of the 11th to 20th weeks'gestation were collected randomly from January 2008 to January 2009. The concentrations of sEng and TGF-β1 in peripheral blood were assayed by ELISA (enzyme-linked immunosorbent assay).Every pregnancy outcome was followed.22 cases with gestational hypertension were included in the study group,176 cases of normal pregnant women were included in control group,2 cases were untouched.Results:1.The serum blood level of the three trimester'sEng in study group was 1.33+ 0.23ng/ml,5.36±0.38ng/ml,6.87±0.25ng/ml, respectively. And the serum concentration of sEng increased along with gestation age. Difference was statistically significant (P<0.05).2.The serum blood level of the three trimester'sEng in control group was 1.10±0.28ng/ml,3.30±0.62ng/ml,4.88±0.39ng/ml, respectively. And the serum concentration of sEng increased along with gestation age. Difference was statistically significant (P<0.05).3.The serum concentration of sEng in study group of the first trimester was higher than that in control group, difference was statistically significantly (P<0.05).4.The serum concentration of sEng in study group of the sencond trimester was higher than that in control group, difference was statistically significant (P<0.05).5.The serum concentration of sEng in study group of the third trimester was higher than that in control group, difference was statistically significant (P<0.05). 6.The serum concentration of TGF-β1 in study group of the three trimester'in study group was 3.47±0.96pg/ml,9.37±2.83pg/ml,13.13±3.62pg/ml, respectively. The serum concentration of TGF-β1 increased along with gestation age. Difference was statistically significant (P<0.05).7.The serum concentration of TGF-β1 in study group of the three trimester'in control group was 3.36±0.75pg/ml,7.37±1.65pg/ml,9.77±2.21pg/ml, respectively. The concentration of TGF-β1 increased along with the gestation age. Difference was statistically significant (P<0.05).8.The serum concentration of TGF-β1 in study group of the first trimester was higher than that in control group, difference wasn't statistically significant (P>0.05).9.The serum concentration of TGF-β1 in study group of the sencond trimester was higher than that in control group.Difference was statistically significant (P<0.05).10.The serum concentration of TGF-β1 in study group of the third trimester was higher than that in control group.Difference was statistically significant (P<0.05).11.The predictive value of sEng on HDCP in the three trimesters was 1.2ng/ml, 4.2ng/ml,5.9ng/ml, respectively, and the predicted value of TGF-β1 on HDCP in the three trimesters was 8.0pg/ml,10.9pg/ml, respectively.12.The serum concentration of sEng was highly correlated with that of TGF-β1(r=0.42,0.56,0.63).Difference was statistically significant (P<0.05).13. The sensitivity and specificity of the predictive value of sEng and TGF-βin the diagnoses of HDCP were 94.5%,97.5%, respectively.Conclusion:1.The serum concentration of sEng increased along with gestation age.2.The serum concentration of TGF-β1 increased along with gestation age.3.The serum concentration of sEng in study group of the first trimester was higher than that in control group, difference was statistically significant (P<0.05).4.The serum concentration of TGF-β1 in study group of the first trimester was higher than that in control group, difference wasn't statistically significant (P>0.05).5.The serum concentration of sEng and TGF-β1 in study group of the sencond and third trimester were obviously higher than that in control group, difference was statistically significant (P<0.05). Section B The study on correlation of TGF-β1 and Endoglin in placenta and myometrium with serum soluble endoglin and TGF-β1Objective:To investigate the TGF-β1, Endoglin in placenta and myometrium and soluble endoglin and TGF-β1 in pregnancy-induced HDCP patients.To explore the correlation of TGF-β1.Endoglin and sEng with pathogenesis of HDCP.Methods:110 cases pregnant women were collected randomly from April 2008 to April 2009.90 cases with HDCP(30 with gestational hypertension,30 with mild pre-eclampsia and 30 with severe pre-eclampsia) in study group.20 normal pregnant women in control group. The serum concentrations of sEng and TGF-β1 were assayed by ELISA.The expression of Eng and TGF-β1 in placenta and myometrium were detected by Immunohistochemistry (IHC).Results:1.The serum concentration of sEng in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 2.93±0.40ng/ml,3.43±0.63ng/ml,5.94±0.53ng/ml,8.23±1.04ng/ml, respectively. And the serum concentration of sEng increased along with gestation age.Difference was statistically significant (P<0.05).2.The serum concentration of TGF-β1 in control group, pregnancy-induced hypertension disease group,mild pre-eclampsia group and severe pre-eclampsia was 9.85±3.19pg/ml,11.21±2.32pg/ml,14.32±4.37pg/ml.15.37±4.93pg/ml, respectively. And the serum concentration of TGF-β1 increased along with gestation age. Difference was statistically significant (P<0.05).3.Positive rate of Endoglin in placenta in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 10.00%,56.70%,86.70%,80.00%.The trend has gradually increased.Diffenrence was statistically significant (χ2=35.43,P<0.01).4. Positive rate of Endoglin in myometrium in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 10.00%,60.00%,86.70%,93.30%.The trend has gradually increased, the difference was statistically significant (χ2=44.89,P<0.01).5.Positive rate of placenta TGF-β1 in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 20.00%,60.00%,86.70%,100.00%, respectively. The trend has gradually increased.The difference was statistically significant (χ2=42.77,P<0.01).6.Positive rate of myometrium TGF-β1 in control group, pregnancy-induced hypertension disease group, mild pre-eclampsia group and severe pre-eclampsia was 10.00%,60.00%,86.7%,96.70%, respectively.The trend has gradually increased.The difference was statistically significant (χ2=48.07,P<0.01).7.The serum concentration of sEng was highly correlated with that of TGF-β1(r=0.41,0.56,0.69).Difference was statistically significant (P<0.05).8.The serum concentration of sEng was highly correlated with the expression of Eng in placenta(r=0.61,0.45,0.37);the serum concentration of sEng was highly correlated with the expression of Eng in myometrium(r=0.74,0.62,0.40). Difference was statistically significant (P<0.05).9.The serum concentration of TGF-β1 was highly correlated with the expression of TGF-β1 in placenta(r=0.59,0.47.0.36);the serum concentration of TGF-β1 was highly correlated with the expression of TGF-β1 in myometrium(r=0.64,0.50,0.39). Difference was statistically significant (P<0.05).10.The expression of TGF-β1 in placenta was highly correlated with the expression of Eng in placenta(r=0.52,0.55,0.60).The expression of TGF-β1 in myometrium was highly correlated with the expression of Eng in myometrium (r=0.53,0.54,0.58).Difference was statistically significant (P<0.05).Conclusion: 1.The expression of sEng in placenta and myometrium in study group was higher than that of control group significantly. 2.The expression of TGF-β1 in placenta and myometrium in study group was higher than that of control group significantly.3.The concentration of serum sEng in pregnancy-induced hypertension group, mild pre-eclampsia group and severe pre-eclampsia group increased obviously,and was highly correlated with the concentration of Eng in placenta and myometrium. The concentration of serum TGF-β1 in pregnancy-induced hypertension group, mild pre-eclampsia group and severe pre-eclampsia group increased obviously.and was highly correlated with the concentration of TGF-β1in placenta and myometrium.4.The concentration of serum sEng of umbilical cord blood was too low to be assayed.5.The concentration of serum TGF-β1 of umbilical cord blood was too low to be assayed.
