The primary study of EphrinB2/EphB4 during the alveolar bone resorptionBackground:Periodontitis is a chronic inflammatory disease, which is characterized by alveolar bone absorption, and alveolar bone repair is currently the greatest problem to cure periodontitis. Thus study the molecular mechanism of bone remodeling is of great practical significance. Qsteoblasts, osteoclasts and their communication are critical in the procedure of bone absorption - formation, but the specific mechanism remains unclear. Recent studies suggest that EphrinB2/EphB4 signaling system may participate in the interaction of osteoblast and osteoclast which may regulate bone homeostasis. However.there is still no report about their effect on alveolar bone resorption during periodontitis.Objective:To improve the technique of experimental Sprague Dawley (SD) rat model with E.coli Lipopolysaccharide (E-LPS) induced alveolar bone resorption, and to explore the potential effect of EphrinB2 and EphB4 in the alveolar bone resorption.Methods:An improved experimental rat model with alveolar bone resorption was established through four times' fixed-point injections with 20μg/μ1 E-LPS. The effect of established model were estimated through histological observation, tartrate resistant acid phosphatase(TRAP) staining, and TRAP stained osteoclast count of the tooth-periodontal samples. Expression of EphrinB2 and EphB4 were detected immunohistochemical method and immunofluorescent test.Results:There are obvious alveolar bone resorption and TRAP stained osteoclasts in the samples of E-LPS induced SD rat model,and the resorption continues at least 20 days. EphrinB2 were significantly noticed in the osteoblasts and osteoclasts, and more EphB4 was shown in the osteogenic zone than the bone absorption area.Conclusions:It is stable and reliable to establish the rat model of alveolar bone resorption with the improved method. EphrinB2 and EphB4 participate in the regulating of alveolar bone resorption. The combined orthodontic-periodontal treatment of periodontitisBackground:Periodontitis could lead to proclination, rotation, overeruption, migration of teeth and traumatic occlusion. The malocclusion can sometimes hinder periodontal treatment and impair function and aesthetics of the stomatognathic system. Traumatic occlusion may inhibit bone repair that can occur following periodontal treatment. In theory, on the basis of well controlled periodontal inflammation, performing orthodontic treatment to rehabilitate aforementioned clinical situations, can not only reestablish good occlusion, but also offers great possibilities for periodontal repair and health maintenance. The combined orthodontic-periodontal treatment has the potential to be a kind of effective treatment mode for periodontitis recommended in clinic.Objective:To observe the therapeutical effect of combined orthodontic-periodontal treatment for patients with periodontitis and malocclusion.and evaluate the effct of this treatment on compromised periodontium.Methods:3 tipical cases of severe periodontitis who have undergone combined orthodontic-periodontal treatment were reported. The orthodontic treatment were given on the basis of the well controlled periodontal inflammation after the initial therapy, then perform periodontal examination and cleaning regularly, and permanently retain the teeth. Dentition, Periodontal indices,and X-ray analysis were evaluated before and after treatment.ResuIts:The malocclusions were corrected and satisfactory occlusion were established after the combined orthodontic-periodontal treatment.There was no progression of periodontal lesions, but improved periodontal indices and alveolar bone repair in certain positions.Conclusions:The combined orthodontic-periodontal treatment is effective for periodontitis, as it can correct the malocclusion and occlusal traumatism.reestablish occlusion,and to some extent promote the repair of the periodontium.
