| ObjectiveThe World Health Organization currently estimates that about 10% to 15% of reproductive couple face low fertility worldwide. Male factors accounted for about 49 percent, among them azoospermia disease is one of the important reasons. Intra-cytoplasmic sperm injection(ICSI) can used to have offspring for the patients whose testicular spermatogenic function was mild inhibited, But the risk of technology itself is cannot be ignored; For the patients who no sperm production or sperm rarely, there is no effective treatment methods at present. How to make azoospermia patients get genetics offspring become clinical research issues that need to be resolved.Protein interacting with Ca kinase 1 (PICK1)was discovered in 1995, It interacts with variety of proteins and performs several functions. It widely distribute in various tissues of the body, particularly in the brain and the testis. The PICK1 protein contains a PDZ structure domain and a BAR structure domain, the PDZ structure domain can combine with many kinds of membrane proteins, and BAR structure domain combine with lipid molecules (predominantly phosphoric acid inositol), through this mechanism PICK1 adjusts the sub-cellular localization and membrane expression of related proteins. The interaction between PICK1 and its target proteins is mediated by the combination of PDZ structure domain and ligand C end, which make the PDZ structure domain become a potential drug targets. Therefore, we can use the specific combination of the PDZ structure domain and small biomolecules to interference or block the natural interaction between PICK1 and ligand protein, and finally achieve the purpose of treatment related diseases. Now the research for PICK1 main study of the mechanism in the nervous system, such as involved in learning and memory, participate in neurohumoral transmission and adjust, and so on. So far, the study on other system has not been carried out in depth. Recent research has shown that PICK1 deficiency causes male infertility in mice by disrupting acrosome formation, The knockout PICK1 mice were found volume of testicular reduced, sperm acrosome deformity and meanwhile born fine tubular apoptosis increased.Based on this, we have designed this research to study the relationship between the relative quantity of PICK1 expression and the spermiogenesis, the quantity expression of testicular tissue of azoospermia with different pathologically diagnosed and its differences were analyzed. Through observe the relationship between the express strength of PICK1 and the rates of fertilization, cleavage, good embryos and clinical pregnancy to preliminary discute its correlation with male fertility.Methods1.50 azoospermia who come our reproductive center from 2007 to 2009, testicular volume≥12 ml, excluding chromosome abnormality, According pathological results and Johnson criteria they were divided inio four groups: SCOS(n=14),MA(n=12),HS(n=9),OA(n=15). The quantity of expression of PICK 1 was tested by imrnunohisto-chemistry integral optical density of the computer graphies analysis system.2. The azoospermia who were diagnosed HS and OA treated by ICSI. Analyzed the rates of fertilization, cleavage, good embryos and clinical pregnancy between the two groups, disscute the relationship between PICK1and male fertility.Results:1. The quantity expression of PICK1:the integrated optieal density values of PICK1 in four groups werel.91±1.01,106.73±8.70,127.79±4.94,147.67±7.48; There was statistically significant difference between them (P<0.05);2. The relationship between and male fertility. There were significant differences on the rates of fertilization, between the two groups (69.1% vs 82.8%, P<0.05); The cleavage, good embryos and clinical pregnancy of them showed no statistically significant differences (68.8% vs 72.1%; 40.9% vs 45.5%; 20.00% vs 36.8%, P >0.05. Conclusion1. The down-regulated quantity expression of PICK1 in the testicular tissue of different pathologically diagnosed may be one of the causes of differentiation dysfunction in the azoospemia patients.2. The quantity expression of PICK 1 may be related with male fertility... |
